“…In a recent review, Tagliaro et al (1989) concluded that separation by reversed-phase HPLC with ECD, because of its specificity, reliability, and sensitivity, is the method of choice for assaying morphine. A coulometric three-electrode arrangement is frequently used because it allows the removal of interfering substances from mobile phase and sample, by selective oxidation at the first two electrodes ( h n d o r f and Kaltenbach, 1986;Svensson, 1986;Kouishi and Hashimoto, 1990;McLean et al, 1990). However, because 5-HT and 5-HIAA are easily oxidized at low potentials at the first electrode and no longer detected at the analytical cell, we in fact could not fully exploit the advantages of the coulometric tluee&xtmde array in our assay.…”

“…In the early 1980s, Derendorf et al measured the drug concentrations in saliva as a proxy of free tissue concentrations and analgesic efficacy 1 before expanding their work to other target phases, such as joints, 2 the brain, 3 and various biological fluids. 4 His group also became interested in local treatment strategies Schreier. [5][6][7] A technological breakthrough was achieved in Hartmut's laboratory in the mid 1990s 8 by adopting the microdialysis technique, as it provided access to unbound interstitial space fluid concentrations.…”

Most of these methods require several timeconsuming sample preparation steps before quantitation.…”
