CRISPR interference and activation of the microRNA-3662-HBP1 axis control progression of triple-negative breast cancer

Abstract: MicroRNA-3662 (miR-3662) is minimally expressed in normal human tissues but is highly expressed in all types of cancers, including breast cancer. As determined with The Cancer Genome Atlas dataset, miR-3662 expression is higher in triple-negative breast cancers (TNBCs) and African American breast cancers than in other breast cancer types. However, the functional role of miR-3662 remains a topic of debate. Here, we found that inhibition or knockout of endogenous, mature miR-3662 in TNBC cells suppresses prolife… Show more

“…Wnt signalling plays an important role in embryonic development, whose deregulation leads to the initiation and progression of human malignancy via regulation of target gene expression. Canonical Wnt/β-catenin signalling is a potential therapeutic target in malignancies, such as NSCLC and triplenegative breast cancer [23]. Malignant tumour cells acquire invasive-associated phenotypes via regulation of the Wnt pathway.…”

“…Proliferation and cell cycle progression of these malignant tumors and their therapy response are also modulated by CRISPR/Cas9 system. [121][122][123][124][125][126][127][128] Noteworthy, delivery of CRISPR/ Cas9 system can pave the way for improving its efficacy in gene expression regulation, which is the focus of the current section. the purpose of breast cancer therapy.…”

“…In macrophage cell lines, CRISPR-mediated indel formation in miR-155 reduced pro-inflammatory cytokine expression in vitro, creating an avenue for treating the autoimmune disease rheumatoid arthritis 103 . miRNA targeting has also been extensively studied in cancer 104 ; various approaches have been shown to lead to reductions in cancer cell proliferation 105 107 , inhibition of metastasis 105 , 106 and death of cancer cells 108 in preclinical studies.…”

“…For example, in settings in which a lncRNA silences a paternal allele, leaving only a mutant maternal allele, the generation of indels in the lncRNA can disrupt its function, leading to the therapeutic expression of the wild-type paternal allele 91,92 . f | To increase the concentration of a therapeutically relevant gene, Cas proteins can be used to form indels in the microRNA (miRNA) or the 3′ untranslated region, leading to a reduction in miRNA binding and amelioration of the resulting RNA interference [101][102][103][104][105][106][107][108] .…”
