CSF isoprostane levels are a biomarker of oxidative stress in multiple sclerosis

Abstract: Objective:To investigate the potential of 8-iso-prostaglandin F2α (8-iso-PGF2α) as a biomarker for disease activity and oxidative stress in the CSF of patients with multiple sclerosis (MS).Methods:The isoprostane 8-iso-PGF2α is an established biomarker for in vivo oxidative stress and lipid peroxidation. We measured CSF 8-isoPGF2α levels in 231 patients with MS (74 with relapsing-remitting MS, 67 with primary progressive MS, and 90 with secondary progressive MS [SPMS]) and 40 controls using a competition ELISA… Show more

“…Some isoprostanes have found clinical applications in obesity, ischemia-reperfusion injury, the central nervous system, cancer, and genetic disorders (Milne et al, 2015). CSF isoprostane levels have been beneficial in evaluating oxidative stress in multiple sclerosis in humans (Mir et al, 2014) whereas plasma or milk isoprostane levels were diagnostic for oxidative stress in lactating dairy cows (Vernunft et al, 2014).…”

“…However, an increase in the CSF concentration of 8-iso-15( S )-PGF 2α was reported in the CSF of patients with MS with F 2 -IP determined by ELISA. 26 28 We attribute these apparent discrepancies to differences in the methods used to determine F 2 -IP. It is well known that results from ELISA are not directly comparable to those determined by mass spectrometry–based assays, 29 with the former being less specific and commonly yielding higher values for 8-iso-15( S )-PGF 2α than mass spectrometry–based assays.…”
