Backgrounds. Curcumin (CUR) is a low-molecular-weight polyphenolic substance obtained from the tuber part of Curcuma species. Anti-inflammatory and anti-hepatitis C virus (HCV) activities have been associated with CUR. However, its poor aqueous solubility and low systemic bioavailability have been the challenges in improving the therapeutic efficacy of curcumin. Aim. The study aimed to produce CUR-loaded liposomal solid gels as anti-HCV delivery systems. Parameters including the physical characteristics and the cell cytotoxicity properties were evaluated. Methods. The freeze-drying technique was applied to manufacture the CUR-loaded liposomal solid gels. Scanning electron microscopy (SEM), X-ray diffractometry (XRD), and differential thermal analysis (DTA) were involved to reveal the characteristics of the solid gels. Such characteristics were as follows: the morphology and the microscopic structure of the solid gels, the crystallinity structure of the curcumin, and the thermal properties of the mixtures. Furthermore, their cell cytotoxicity was investigated using a Huh7it cell line. Results. The SEM images confirmed that curcumin liposomes were intact and trapped in the solid gel matrix. The XRD data showed flat patterns diffractograms of the formulations, confirming the transformation of CUR from crystalline to amorphous form. The DTA thermograms showed a single melting endothermic peak at a higher temperature around 200°C, indicating a single-phase transition of the mixtures. The XRD and DTA data revealed the molecular dispersion of CUR in the developed formulations. The cytotoxicity data provided as cell cytotoxicity 50 (CC50) for all formulations were ≥25 mg. These data confirmed that the developed liposomal solid gels were not cytotoxic to Huh7it cell line, indicating that the anti-HCV activity would be through a specific pathway and not by its toxicity. Conclusion. The CUR-loaded liposomal solid gels exhibited the potential and offered an alternative dosage form to improve the therapeutic efficacy of curcumin as an anti-HCV.

Preparation of Curcumin-Eudragit® E PO Solid Dispersions with Gradient Temperature through Hot-Melt Extrusion

Hot-melt extrusion (HME) has great advantages for the preparation of solid dispersion (SD), for instance, it does not require any organic solvents. Nevertheless, its application to high-melting-point and thermosensitive drugs has been rarely reported. In this study, thermally unstable curcumin (Cur) was used as a drug model. The HME process was systematically studied by adjusting the gradient temperature mode and residence time, with the content, crystallinity and dissolution of Cur as the investigated factors. The effects of barrel temperature, screw speed and cooling rate on HME were also examined. Solubility parameters and the Flory–Huggins method were used to evaluate the miscibility between Cur and carriers. Differential scanning calorimetry, X-ray diffraction, Fourier transform infrared spectroscopy, equilibrium solubility and in vitro and in vivo experiments were used to characterize and evaluate the results. An amorphous Cur SD was successfully obtained, increasing the solubility and release of Cur. In the optimal process, the mass ratio of Cur to Eudragit® E PO (EPO) was 1:4 and the barrel temperature was set at a gradient heating mode (130 °C–135 °C–140 °C–145 °C–150 °C–155 °C–160 °C) at 100 rpm. Related pharmacokinetic test results also showed the improved bioavailability of the drug in rats. In a pharmacodynamic analysis of Sprague–Dawley rats, the Cmax and the bioavailability of the Cur-EPO SD were 2.6 and 1.5 times higher than those of Cur, respectively. The preparation of the amorphous SD not only provided more solubility but also improved the bioavailability of Cur, which provides an effective way to improve the bioavailability of BCS II drugs.

The Influence of Electrode Plate Area in Reducing Turbidity and Color Levels in Peat Water

Peat water generally exhibits characteristics ranging from dark brown to black, with high organic content and a pH range between 3.7 and 5.3. These features pose challenges in meeting clean water quality standards, essential for maintaining human health. Consequently, the treatment of peat water becomes a pressing need to enhance its quality for suitable use. Electrocoagulation has emerged as a widely applied method in peat water treatment. This research aims to evaluate the influence of electrode plate area and contact time in reducing turbidity and color in peat water. The research methodology employed a quasi-experimental design known as the One-Group Pretest-Posttest. Thirty test samples were collected with variations in electrode plate areas of 4 x 30 cm, 5 x 30 cm, and 6 x 30 cm. Contact time was varied within the range of 30, 60, 90, 120, and 150 minutes, applying a voltage of 48 volts. The results indicated fluctuations in the increase of turbidity in peat water after the electrocoagulation treatment. A plate area of 6 x 30 cm led to a turbidity increase of 27.33 NTU or approximately 83% after a contact time of 150 minutes. Meanwhile, the color level in peat water decreased by 65 TCU with a 5 x 30 cm electrode plate area after 150 minutes of contact time, achieving an effectiveness of 90% in color reduction. In conclusion, the electrode plate area significantly influences the reduction of turbidity and color in peat water. Although there is no discernible effect of contact time on turbidity levels, prolonged contact time has a significant impact on reducing the color level in peat water.
