Current Landscape in Organic Nanosized Materials Advances for Improved Management of Colorectal Cancer Patients

Abstract: Globally, colorectal cancer (CRC) ranks as one of the most prevalent types of cancers at the moment, being the second cause of cancer-related deaths. The CRC chemotherapy backbone is represented by 5-fluorouracil, oxaliplatin, irinotecan, and their combinations, but their administration presents several serious disadvantages, such as poor bioavailability, lack of tumor specificity, and susceptibility to multidrug resistance. To address these limitations, nanomedicine has arisen as a powerful tool to improve cu… Show more

“…Several combination chemotherapeutic regimens such as FOLFIRI (folinic acid, 5-FU and IRI), XELOX/CAPOX (OXA and capecitabine) and FOLFOX (folinic acid, 5-FU and OXA) are widely used in clinical treatment for CRC[ 10 ]. However, and despite the continued efforts devoted to advance CRC chemotherapy, the prognosis of this disease is still unsatisfactory, particularly for metastatic CRC patients[ 11 ].…”

“…The use of polymer nanoaggregates as carriers for drug delivery revolutionized pharmacology and opened up new possibilities for an extended circulation time, reduced toxicity, optimized release of active substances, and targeted drug delivery. [1][2][3] Polymer nanoaggregates loaded with anticancer drugs have been already successfully used for treatment of various types of cancer [4] and can eventually lead to an effective therapy for his disease which killed nearly 10 million people only in 2020. [5] Doxorubicin [6,7] and paclitaxel [8,9] are among the drugs with a broad anticancer activity, which, in some cases show a synergetic effect when used together.…”

“…Chemotherapy and surgery are the main treatment for CAC clinically. Fluorouracil and molecular targeted drugs are clinical drugs but with many adverse reactions ( Fiala et al, 2019 ; Ginghină et al, 2021 ). At the same time, there is a possibility of complications in the above treatments, and long-term medication will also reduce the compliance of patients.…”
