Traditional methods for the assessment of dietary intake are prone to error; in order to improve and enhance these methods increasing interest in the identification of dietary biomarkers has materialised. Metabolomics has emerged as a key tool in the area of dietary biomarker discovery and to date the use of metabolomics has identified a number of putative biomarkers. Applications to identify novel biomarkers of intake have in general taken three approaches: (1) specific acute intervention studies to identify specific biomarkers of intake; (2) searching for biomarkers in cohort studies by correlating to self-reported intake of a specific food/food group(s); (3) analysing dietary patterns in conjunction with metabolomic profiles to identify biomarkers and nutritypes. A number of analytical technologies are employed in metabolomics as currently there is no single technique capable of measuring the entire metabolome. These approaches each have their own advantages and disadvantages. The present review will provide an overview of current technologies and applications of metabolomics in the determination of new dietary biomarkers. In addition, it will address some of the current challenges in the field and future outlooks.

Metabolomics is an important "omics" technology, complementary to genomics and proteomics, as parts of systems biology, giving information (qualitative fingerprints and quantitative profiling) as a mirror of cell and extracellular metabolic activity. A cohort of small metabolites are involved in the control and regulation of cellular functions, as intermediates or final products, their presence or levels being useful for the early diagnosis of different pathologies. Bioinformatics tools are mandatory for a future "computational" metabolomics, needed to manage large number of experimentally acquired data obtained from biological samples (plants, animal or human tissues). This review presents updated information about different high-throughput analytical techniques and data acquisition software (1-2), the pre-processing of data, converted to specific matrices, further processed by specific normalization and alignment procedures (3), then analysed by statistical univariate and multivariate chemometric and /or statistical techniques (4), identifying biomarkers by comparison with databases (5), and finally elucidating the networks and pathways (6). New software is available for data conversion, pre-processing, alignment algorithms, bucketing, normalization, underlying the challenges and comparisons with international data bases. Finally, the accurate identification of individual molecules as biomarkers, either evaluated by untargeted metabolomics techniques (Principal Component Analysis -PCA), Cluster Analysis -CA) or supervised ones (Partial Least Square Discriminant Analysis (PLS-DA) is presented. The accurate identification of metabolites and their involvement in metabolic networks and pathways became possible by well-established databases (HMDB, LIPID MAPS, KEGG, etc.), to validate all experimental data. Bioinformatics is a sine-qua-non tool, to be used and valorised by untargeted or targeted metabolomics, as an integrated technology in systems biology.

ResumoO câncer de bexiga é uma neoplasia urogenital que acomete homens e mulheres, sendo que somente no Brasil 8.600 novos casos ao ano são diagnosticados. Cistoscopia transuretral é a conduta padrão no diagnóstico e acompanhamento do câncer de bexiga. Entretanto, tal procedimento é extremamente invasivo e doloroso além de ter elevado custo e não garantir todos os resultados. Assim, busca-se por marcadores moleculares que possam auxiliar no diagnóstico e progressão do câncer de bexiga, bem como diminuir a necessidade de exames invasivos no acompanhamento de pacientes tratados. Nesse sentido, a urina tem papel de destaque como fonte de biomarcadores devido principalmente ao seu caráter não invasivo.Nesse trabalho foram utilizadas duas abordagens 'ômicas': proteômica e metabolômica, para a busca de biomarcadores em urina para o diagnóstico e prognóstico do câncer de bexiga, respectivamente. Com a abordagem proteômica buscou-se apenas por biomarcadores para o diagnóstico da doença e, utilizando as técnicas de eletroforese 2-DE, OFFGEL e MS, juntamente com análise estatística multivariada, foi possível identificar 32 proteínas que apresentam-se como potenciais marcadores para o câncer de bexiga. A abordagem metabolômica foi empregada para a busca de biomarcadores para reincidência e progressão da doença. As técnicas analíticas utilizadas nessa abordagem, LC-MS e CE-MS, mostraram-se complementares e, dos resultados obtidos com ambas e avaliados com análise estatística multivariada foi possível indicar 19 metabólitos para reincidência e 23 metabólitos para progressão do câncer de bexiga.Assim, neste trabalho explorou-se como as ciências 'ômicas', a qual abrange técnicas analíticas de high-throughput, estatística multivariada e ferramentas de bioinformática auxiliando na descoberta de potenciais biomarcadores não invasivos para o diagnóstico e prognóstico do câncer de bexiga.
