Cyclic Pressure Stimulates DNA Synthesis through the PI3K/Akt Signaling Pathway in Rat Bladder Smooth Muscle Cells

Abstract: Previous studies demonstrated that the bladder exhibited severe tissue remodeling following spinal cord injury. In such pathological bladders, uninhibited non-voiding contractions subject bladder cells to cyclic oscillations of intravesical pressure. We hypothesize that cyclic pressure is a potential trigger for tissue remodeling in overactive bladder. Using a custom-made setup, rat bladder smooth muscle cells (SMC) in vitro were exposed to cyclic hydrostatic pressure (40 cm H2O) at either 0.1 Hz or 0.02 Hz fr… Show more

“…AoSMCs were treated with vehicle control (dimethyl sulfoxide), 100 nmol/L rapamycin, 26 10 mol/L UO126, 27 or 20 mol/L LY-294002. 28 Rapamycin, U0126, and LY294002 were purchased from LC Laboratories (Woburn, Mass). The cell proliferation rate was determined with use of the WST-1 cleavage-based cell proliferation (Chemicon International, Temecula, Calif) as described.…”

“…The surmounting evidence of bladder SMC mechanotransduction includes, but is not limited to, increased expressions of heparin-binding epidermal growth factor (HB-EGF) and cyclooxygenase-2 (COX-2) 15,17-19 and activation of p38 SAPK2 and PI3K/Akt intracellular signaling pathways. 2,15,23 These results suggest that multiple signaling pathways may be concomitantly at work in response to the applied mechanical stimuli. Thus, the conventional reductionist approach that examines only one or two molecules at a time may not fully uncover the complexity of the cellular mechanotransduction events.…”

“…1). 23 SMC maintained under static (atmospheric pressure) conditions for the duration of the experiment were used as the control. This set of parameters was chosen because in our previous study the earliest molecular response by SMC to the cyclic pressure was detected at the 1 h time point.…”
