Cytocompatibility and cell migration evaluation of calcium silicate‐based root canal sealer compared to epoxide‐amine resin sealer in stem cells from human apical papilla: An in‐vitro study

Abstract: The purpose of this study was to assess the effect of a calcium silicate‐based sealers (CeraSeal) and an epoxy resin‐based sealer (AH Plus) on cytotoxicity and cell migration of stem cell from the human apical papilla (hSCAPs) by using the Alamar Blue, Annexin V‐FICT and wound healing assays. In Alamar Blue assay, hSCAPs exposed to undiluted CeraSeal extract had significantly higher cell viability compared with that observed when cells were treated with AH Plus in all experimental period (p < 0.001). The fl… Show more

“…In fact, to the author’s knowledge, this is the first study on BrF and, to date, there is scarce evidence on the biological properties of AHPbcs and its immunomodulatory potential yet to be elucidated [ 25 , 27 , 30 , 47 ]. Parallelly, AHP was used as a comparison, given its extensive evidence and common use as a control in similar studies [ 26 , 48 , 49 ].…”

“…Regarding cellular migration, literature suggests that the mechanisms underlying wound closure in the scratch assay encompass both cellular proliferation and migration capabilities [ 20 ]. Indeed, the interaction between biomaterials and cellular structures can precipitate cellular degeneration and impede the process of wound healing [ 21 ].…”

“…Indeed, the interaction between biomaterials and cellular structures can precipitate cellular degeneration and impede the process of wound healing [ 21 ]. In the context of this study, cells treated with bioceramics exhibited responses analogous to those observed in the untreated control group throughout the duration of the wound healing assay [ 20 , 22 ]. Furthermore, an initial assessment of cytotoxicity was conducted through a morphological analysis utilizing immunofluorescence techniques.…”

“…It has been described that the wound healing closure in the scratch assay involves proliferation and migration capacity (Nguyen et al, 2023). In fact, biological interactions between biomaterials and cells can cause cellular degeneration and delay wound healing (Sanz et al, 2021).…”
