Streptomyces sp. MUM273b: A mangrove‐derived potential source for antioxidant and UVB radiation protectants

Microbial natural products serve as a good source for antioxidants. The mangrove‐derived Streptomyces bacteria have been evidenced to produce antioxidative compounds. This study reports the isolation of Streptomyces sp. MUM273b from mangrove soil that may serve as a promising source of antioxidants and UV‐protective agents. Identification and characterization methods determine that strain MUM273b belongs to the genus Streptomyces. The MUM273b extract exhibits antioxidant activities, including DPPH, ABTS, and superoxide radical scavenging activities and also metal‐chelating activity. The MUM273b extract was also shown to inhibit the production of malondialdehyde in metal‐induced lipid peroxidation. Strong correlation between the antioxidant activities and the total phenolic content of MUM273b extract was shown. In addition, MUM273b extract exhibited cytoprotective effect on the UVB‐induced cell death in HaCaT keratinocytes. Gas chromatography–mass spectrometry analysis detected phenolics, pyrrole, pyrazine, ester, and cyclic dipeptides in MUM273b extract. In summary, Streptomyces MUM273b extract portrays an exciting avenue for future antioxidative drugs and cosmeceuticals development.

Objective: Microorganisms, especially bacteria and its proteins have proven to be potential anti-cancer agents as they selectively attack the tumor cells or tumor micro-environments. The extract of Pseudomonas aeruginosa found to contain proteins that have shown promising anticancer activity. In this work, it was attempted to increase the biomass and trigger the total protein fraction of Pseudomonas aeruginosa (MTCC 2453). Methods:The organism was cultivated in three different such as Luria-Bertani (LB) broth, minimal medium9 (M9), super broth medium (SB) and asparagine-proline (AP) broth. Asparagine proline broth was selected as it has shown high cell growth rate. The media was further optimized by the addition of NaHCO3 and copper sulphate to trigger the protein production. Optimized Aspergine proline broth has achieved highest cell biomass. After the shake flask culture, the overnight grown culture in optimized AP medium was further grown in a 5 L bioreactor by fed-batch cultivation to achieve higher cell densities. Results:The highest protein production was achieved at 40 ° C. Highest biomass and protein content was observed at pH 8 while lowest biomass was produced at pH 2. A gradual increase in biomass content observed from 12 h towards to 48 h. Conclusion:High biomass and proteins content and of Pseudomonas aeruginosa (MTCC 2453) can be produced in optimized asparagine-proline broth. Further the extract is purified to produce novel anti-cancer proteins.

Pseudomonas spp. have been widely studied for their plant growth promoting and antimicrobial metabolites. The genus got attention due to the production of array of secondary metabolites involved in the suppression of phytopathogens and ability to stimulate plant growth by means of nitrogen-fixation, production of hydrolytic enzymes, regulatory hormones, and solubilization of inorganic minerals. In recent years, research was focused towards identification of biosynthesis pathways and genes involved in the production of secondary metabolites that led to the discovery of novel metabolites including many new phenazine derivatives, quorum-sensing signals, rhizoxin analogues, cyclic lipopeptides, and a new class of alkylsubstituted aromatic acids. Identification of these biosynthetic pathways provided insights for their successful application in agriculture and for environmental sustainability. In addition, many genomic and metabolomic databases such as; METLIN, KEGG, GNPS, CFM-ID, MassBank, and MetaboLights, allowed exploring intricate metabolic pathways and significant genes involved in the biosynthesis of compounds. Several softwares, genome-mining tools and new techniques, such as MALDI-IMS and MALDI-FTICR MS were developed to facilitate the characterization of new metabolites. Additionally, use of MALDI-imaging techniques facilitated real-time visualization of complex microbial communities and their relationship with pathogens. Secondary metabolites of Pseudomonas spp. were also demonstrated for their apoptotic, anti-mitotic, nematocidal, herbicidal, anthelmintic, insecticidal, and phytotoxic effects. Total biosynthesis of metabolic derivatives and genetic engineering enabled to develop strains with improved yield of targeted bio-products. Availability and access to published genomic sequences and comparative bioinformatics tools helped in identification of strain-specific traits and development of multifunctional inocula. This review highlights significant advances in identification of Pseudomonas secondary metabolites for their successful agricultural and pharmaceutical applications.
