An amino-substituted bis(aryldiazomethane) was synthesized and used to modify cotton fibers by a carbene insertion reaction. A subsequent amidation of the modified cotton fibers introduced thioether groups onto the surface. Thus, three different types of cotton fibers with hydroxyl, amine, or thioether groups were expected to bind silver nanoparticles (Ag NPs), which were characterized by scanning electron microscopy to show the surface morphology, as well as X-ray photoelectron spectroscopy, energy-dispersive X-ray spectroscopy, and thermogravimetric analysis to quantify the loading of Ag NPs. A bioassay experiment showed antibacterial activity of the modified cotton fibers. The development of these antibacterial cotton fibers not only gave a clear and deep comparison of different silver-binding approaches but also had potential industrial and daily applications for medical textiles.

Despite the progress of the development of antimicrobial therapeutics, the whole world is still under pressure of several microbial diseases. Antimicrobial drug development is therefore considered as one of the most practicable research works at present time. Even most of the pharmaceutical companies are investing to develop better therapeutic solutions against the life-threatening infectious diseases caused by Mycobacterium tuberculosis, Helicobacter pylori, Vibrio cholerae, Entamoeba histolytica, Plasmodium falciparum and many others. These microbial pathogens are not only a curse for human health but also a result in huge economic losses by affecting the health of economically important animals like poultry, cattle and other livestock. Considering the urgency, several effective antibiotics have been developed to combat microbial diseases and are available in the market. However, emergence of resistance against these drugs due to the maluses has created an alarming situation. In this scenario, the use of bioactive noble metal nanoparticles (silver, gold and platinum nanoparticles) has shown better therapeutic efficiency in terms of low treatment dose, less toxicity and absence of microbial resistance. Moreover, the use of several surface modifiers, coating and stabilizing agents resulted in enhancement of the bioactivity, rapid delivery and controlled drug release, improvement of biocompatibility and cytotoxicity. In this chapter, we have presented a comprehensive overview on the antimicrobial efficacy of noble metal nanoparticles along with the mechanistic insights behind their activity at the cellular and molecular level. Silver nanoparticles as antimicrobial therapeutics: current perspectives and future challenges

Utility of silver metal in antimicrobial therapy is an accepted practice since ages that faded with time because of the identification of a few silver resistant strains in the contemporary era. A successive development of antibiotics soon followed. However, due to an indiscriminate and unregulated use coupled with poor legal control measures and a dearth of expertise in handling the critical episodes, the antibiotics era has already seen a steep decline in the past decades due to the evolution of multi-drug resistant 'superbugs' which pose a sizeable challenge to manage with. Due to limited options in the pipeline and no clear strategy in the forefront, the aspirations for novel, MDR focused drug discovery to target the 'superbugs' arose which once again led to the rise of AgNPs in antimicrobial research. In this review, we have focused on the green routes for the synthesis of AgNPs, the mode of microbial inhibition by AgNPs, synergistic effect of AgNPs with antibiotics and future challenges for the development of nano-silver-based therapeutics.
