Daily Rhythms in the Cyanobacterium Synechococcus elongatus Probed by High-resolution Mass Spectrometry–based Proteomics Reveals a Small Defined Set of Cyclic Proteins

Abstract: Circadian rhythms are self-sustained and adjustable cycles, typically entrained with light/dark and/or temperature cycles. These rhythms are present in animals, plants, fungi, and several bacteria. The central mechanism behind these "pacemakers" and the connection to the circadian regulated pathways are still poorly understood. The circadian rhythm of the cyanobacterium Synechococcus elongatus PCC 7942 (S. elongatus) is highly robust and controlled by only three proteins, KaiA, KaiB, and KaiC. This central clo… Show more

“…Finally, we performed untargeted metabolic profiling of WTcells and ΔkaiC mutants to investigate how oscillator activity affects global metabolite abundance at the transition from darkness into light. We present a hypothesis for clock regulation of diurnal metabolism that combines our data with previous reports on S. elongatus transcript and protein rhythms (17,21,23) and that highlights the importance of circadian output for proper metabolite partitioning under LD growth regimes.…”

“…A few studies have investigated the transcriptome, proteome, and physiological dynamics of particular species of cyanobacteria over a 24-h period under LD growth (6,22,23). In general, systems for oxygenic photosynthesis are activated during the day, and systems for respiratory metabolism are activated at night.…”

“…For example, a major unknown factor is the relative amount of noncatalytic proteins, reported to be up to 55% of total protein for slow-growing cells (31). We hypothesize that, for fast-growing cells, this percentage is considerably lower.…”

“…measurements over a full diurnal cycle have been conducted yet, several studies have investigated the transcriptome, proteome, and physiology of S. elongatus PCC 7942 and other cyanobacteria over a full 24-h diurnal cycle (9,31,33,34). Published transcriptome studies of cyanobacteria typically show global rhythms in gene expression, including a significant reduction of the transcriptional output during the dark period (33,35,36).…”
