Data on preparation and characterization of an insect odorant receptor based biosensor

Abstract: Insect Odorant receptors (OrXs) can be used as the recognition element in a biosensor as they demonstrate high levels of sensitivity and selectivity towards volatile organic compounds. Herein, we describe a method to express and purify insect odorant receptors and reconstitute them into artificial lipid bilayers (liposomes). These OrX/liposomes were covalently attached to a gold surface and characterized using quartz crystal microbalance with dissipation monitoring (QCM-D). The interaction of OrX/liposomes imm… Show more

“…Out of the seven responsive OR cell lines reported here only one, OR13a was stably expressed so far in an insect cell line (Sf21) 35 . This and other ORs were expressed transiently either by mRNA injection into Xenopus oocytes or by transfection into immortalized cell lines: OR13a 36 , OR10a and OR71a 37 , 38 , OR47a 9 , 34 , 39 , OR49b 39 , 40 , and OR85b 41 , 42 . In contrast, all our stable cell lines can be used for months for direct measurements.…”

“…Out of the seven responsive biosensor cell lines reported in our study only one, OR13a was previously generated as a stable insect cell line (Sf21; Termtanasombat et al, 2016). Some other ORs were expressed only transiently either by mRNA injection into Xenopus oocytes or by transfection into immortalized cell lines: OR10a and OR71a (Khadka et al, 2018;Murugathas et al, 2019), OR47a (Miazzi et al, 2019;Röllecke et al, 2013;Sato et al, 2008), OR49b (Kolesov et al, 2021;Röllecke et al, 2013), and OR85b (Misawa et al, 2010;Nichols and Luetje, 2010). In contrast, all cellular biosensors generated in this study are stable cell lines that can be used for months for direct measurements.…”

“…The feasibility and effectiveness of such systems using biomolecules as materials to detect odor molecules in liquids have been demonstrated. [ 50a,55 ] In recent years, two new detection methods, localized SPR (LSPR) [ 56 ] and SPRi, [ 57 ] have been proposed based on SPR.…”

“…QCMs have been widely used in the development of various types of B‐ENs. [ 50a,c,64 ] QCMs are more sensitive than SAWs and are capable of detecting concentrations up to the parts per million (ppm) level. [ 65 ] To improve the sensitivity of SAW, a thin layer of diamond nanoparticles can also be coated on the surface of the sensor, detecting odors up to the parts per billion (ppb) level.…”
