DDX6 Helicase Behavior and Protein Partners in Human Adipose Tissue-Derived Stem Cells during Early Adipogenesis and Osteogenesis

Abstract: DDX6 helicase is an RNA-binding protein involved in different aspects of gene expression regulation. The roles played by DDX6 depend on the complexes associated with it. Here, for the first time, we characterize the protein complexes associated with DDX6 in human adipose tissue-derived stem cells (hASCs) and analyze the dynamics of this helicase under different conditions of translational activity and differentiation. The results obtained demonstrated that the DDX6 helicase is associated with proteins involved… Show more

“…A downregulated gene, DDX6 , which encodes DEAD-box helicase 6 protein, has been reported to function in adipogenesis. In human adipose tissue-derived stem cells, the number of DDX6 granules per cell was reduced during adipogenesis [ 42 ].…”

“…hASCs were isolated from adipose tissue obtained from one otherwise healthy female donor (BMI = 27.03) who had undergone liposuction surgery. The isolation protocol followed previously published procedures ( Rebelatto et al, 2008 ; Marcon et al, 2020 ). In short, the tissue was first washed with sterile phosphate-buffered saline (PBS) and then digested with collagenase type I (Gibco ® , Invitrogen ® , Carlsbad, CA, United States) for 30 min at 37°C, 5% CO 2 and under constant agitation.…”

“…NBDY (non-annotated P-body dissociating polypeptide) was initially characterized as a human microprotein component of the mRNA decapping complex, directly interacting with EDC4 and DCP1A being localized in Pbodies' granules [53,57]. It has recently been demonstrated that DCP1A is dispersed in granular structures in the hASC cytoplasm, partially co-located with DDX6 (another interaction partner of NBDY) [53,58]. Another example is the smORF of LINC00116 that encodes the 56-aa microprotein MTLN (Figure 3B).…”
