Deciphering c-MYC-regulated genes in two distinct tissues

Abstract: BackgroundThe transcription factor MYC is a critical regulator of diverse cellular processes, including both replication and apoptosis. Differences in MYC-regulated gene expression responsible for such opposing outcomes in vivo remain obscure. To address this we have examined time-dependent changes in global gene expression in two transgenic mouse models in which MYC activation, in either skin suprabasal keratinocytes or pancreatic islet β-cells, promotes tissue expansion or involution, respectively.ResultsCon… Show more

“…In support of these findings, we observed a marked increase in Igf1 and Igf2 gene expression at gestational day 20 as a result of growth restriction, with a concomitant increase in cardiac Cmyc mRNA expression. There is evidence that variable expression of Igf1 can influence expression of downstream Cmyc in murine skin cells, pancreatic beta-cells (35) and human preadipocytes (14). However, the increased cardiac Bcl2 gene expression we observed at gestational day 20 is contradictory to previous literature, which showed overexpression of Igf1 in mice can attenuate aging-induced increases in cardiac Bcl2 gene expression (23).…”

“…C-myc has been shown to negatively regulate e-cadherin through activating the transcription of microRNA-9 in breast cancer cells [19], which was consistent with our observation. In addition, c-Myc can induce expression of Cyclin D1 to promote cell cycle progression in multiple systems [20,21], and functional interactions between c-Myc and cylclin D1 play a critical role in tumor initiation, progression and response to chemotherapy [22]. Therefore, we suspect that our observation that knockdown of ZXF2 led to down regulation of Cyclin D1 could also be c-Myc dependent.…”

“…3). Of note, Myc is also an oxidative stress-responsive gene (Elouil et al 2005, Jonas et al 2009) that has been implicated in β-cell dedifferentiation (Jonas et al 2001, Laybutt et al 2002c, Pascal et al 2008, Robson et al 2011. On the other hand, it has been proposed that Myc expression is negatively regulated by PDX1 (Chen et al 2007).…”
