Defective lymphoid development in mice lacking expression of the common cytokine receptor γ chain

Abstract: The common gamma chain (gamma c) of the IL-2, IL-4, IL-7, IL-9, and IL-15 receptors is defective in humans with XSCID. Mice lacking gamma c expression had hypoplastic thymuses; the thymocytes responded to gamma c-independent mitogens, but not gamma c-dependent stimuli. Splenic T cells were diminished at 3 weeks of age, but CD4+ T cells markedly increased by 4 weeks. B cells were greatly diminished in contrast with the situation in XSCID. NK cells, gamma delta intestinal intraepithelial lymphocytes, dendritic e… Show more

“…In the null mutant mice, development of IEL is severely diminished and PP are not detected (28,29). We examined extensively tissue sections of small intestine prepared from male mice with a truncated mutant of the CR␥ chain (CR␥ Ϫ/Y mice) that showed a phenotype similar to that of the null mutant mice in terms of development of the lymphocyte population (31,32) and verified that CP were undetectable.…”

“…CD132-null mice (B6.129S4-IL2rg tm1Wjl /J) (17), LT␣ Ϫ/Ϫ mice (B6.129S2-Lta tm1Dch /J) (5), and TNFRI/TNFRII Ϫ/Ϫ mice were purchased from The Jackson Laboratory (Bar Harbor, ME). Breeding colonies of the CD132-null and LT␣ Ϫ/Ϫ mice were maintained in a conventional mouse facility at Emory University.…”

“…Studies of gene-targeted mice have provided much of the evidence implicating specific cytokines in ␣␤ T cell, ␥␦ T cell, or NK cell development. For instance, mice lacking IL-7 or components of the IL-7R signaling pathway (i.e., IL-7R␣, common ␥-chain (␥c), 3 or Jak3) fail to develop ␥␦ T cells (1)(2)(3)(4)(5)(6)(7). IL-15R signaling has been shown to be critical for NK cell development, as IL-15R␣-, ␥c-, and Jak3-deficient mice all lack NK cells (4 -10).…”
