Deleted in Malignant Brain Tumors 1 (DMBT1) is present in hyaline membranes and modulates surface tension of surfactant

Abstract: Background: Deleted in Malignant Brain Tumors 1 (DMBT1) is a secreted scavenger receptor cysteinerich protein that binds various bacteria and is thought to participate in innate pulmonary host defense. We hypothesized that pulmonary DMBT1 could contribute to respiratory distress syndrome in neonates by modulating surfactant function.

“…Indeed, high levels of DMBT1/gp340/SAG were found in the tracheal aspirates and lung tissue sections of newborn infants with bacterial infections (Mü ller et al, 2007) and in the A549 lung cell line after pro-inflammatory stimulation with PMA (Kang et al, 2002). Inflammation-whether induced by TNF␣, by LPS, or by inflammatory bowel disease-also up-regulated DMBT1/gp340/SAG expression in intestinal epithelial cells Rosenstiel et al, 2007).…”

“…The DMBT1 expression in these hypoxic lungs is significantly higher than expected in these gestational ages as demonstrated (Figure 1 C,F) and already described by our research group. 5,6 3.2 | DMBT1 expression in A549 cells after normoxia and hypoxia…”

“…Earlier studies demonstrated that hrDMBT1 is able to inactivate the function of surfactant preparations widely used to treat respiratory distress syndrome. 5 In this study we analyzed the effect of hrDMBT1 on the surfactant ultrastructure as the morphology of surfactant is closely related to its function. 11 Lamellar body-like forms are regarded as surface-active precursors from which the active surface layer is generated.…”
