Dendritic cells with TGF-β1 differentiate naïve CD4+CD25T cells into islet-protective Foxp3+regulatory T cells

Abstract: CD4 ؉ CD25 ؉ Foxp3 ؉ regulatory T cells (T regs) are important for preventing autoimmune diabetes and are either thymic-derived (natural) or differentiated in the periphery outside the thymus (induced). Here we show that ␤-cell peptide-pulsed dendritic cells (DCs) from nonobese diabetic (NOD) mice can effectively induce CD4 ؉ CD25 ؉ Foxp3 ؉ T cells from naïve islet-specific CD4 ؉ CD25 ؊ T cells in the presence of TGF-␤1. These induced, antigen-specific T regs maintain high levels of clonotype-specific T cell r… Show more

“…DC can differentiate T regs in the thymus [78] and in the periphery [79], and expand them once the T regs have formed [80][81][82]. Antigen-specific T regs are much more effective than polyclonal populations in suppressing immunity in vivo [79,[81][82][83]. For many years, the major approach to the induction of peripheral tolerance has been to inject very large doses of antigen.…”

“…Mice lacking Stat3 in myeloid compartment of tumor stroma, including DCs and macrophages, showed reduced numbers of tumor-infiltrating CD4 + CD25 + /Foxp3 + /Lag-3 + Tregs, which was accompanied by increased CD8 + effector T cells. An important role of TGFβ in mediating Treg expansion through direct interaction of CD4 + CD25 + cells with DCs has been demonstrated in various systems [20,21]. In addition, resident tumor DCs might also actively promote immunosuppression through expansion of Tregs [22].…”

“…Most studies have focused on dendritic cells (DC), which are thought to be key players in generating peripheral Treg [20][21][22]. B cells are also APC that present antigen to T cells.…”
