Deregulated Gab2 phosphorylation mediates aberrant AKT and STAT3 signaling upon PIK3R1 loss in ovarian cancer

Abstract: Copy number loss of PIK3R1 (p85α) most commonly occurs in ovarian cancer among all cancer types. Here we report that ovarian cancer cells manifest a spectrum of tumorigenic phenotypes upon knockdown of PIK3R1. PIK3R1 loss activates AKT and p110-independent JAK2/STAT3 signaling through inducing changes in the phosphorylation of the docking protein Gab2, thereby relieving the negative inhibition on AKT and promoting the assembly of JAK2/STAT3 signalosome, respectively. Additional mechanisms leading to AKT activa… Show more

“…PIK3R1 has been identified to be differentially expressed in many human cancers. For example, PIK3R1 functions as a tumor suppressor in hepatocellular carcinomas and renal cancer [27,28], whereas acts as an oncogene in ovarian and colon tumors and plays a role in tumor progression and metastasis [29,30]. However, the relationship between PIK3R1 and LSCC cell development has not been fully elucidated.…”

“…Although there is functional specificity among p110 isoforms 1,2 , the roles of p85 isoforms in cancers are only beginning to be understood. PIK3R1 (encodes p85α) has been suggested to act as a tumor suppressor through functions such as inhibiting p110 kinase activity and stabilizing phosphatase and tensin homolog (PTEN) 3,4 . Depletion of p85α can thus lead to enhanced p110 activity and PTEN destabilization, as well as cell context-dependent activation of oncogenic signaling [3][4][5] .…”

Depletion of p85α can thus lead to enhanced p110 activity and PTEN destabilization, as well as cell context-dependent activation of oncogenic signaling [3][4][5] . Indeed, loss-of-function disruptions in PIK3R1 are frequent in cancers, including copy number loss and truncation or point mutations.…”

“…Our findings show that the expression of both SOCS1 and SOCS3 show a high degree of mutual exclusivity with PIK3R1 ( Figure 6A). Even though this gene codes for the p85 regulatory subunit of PI3K, there is strong evidence indicating its role also as a tumor suppressor by positively modulating PTEN, a negative regulator of the PI3K pathway, and by modulating AKT and STAT3 activation [101][102][103]. Consistent with this, high PIK3R1 predicts favorable survival, whereas its lower expression in synergy with low SOCS1 predicts poor survival ( Table 2).…”
