Dermasence refining gel modulates pathogenetic factors of rosacea in vitro

Abstract: We show evidence explaining the anti-inflammatory effect of Dermasence Refining Gel in rosacea pathogenesis in vitro. The adjunctive use of DRG in mild to moderate rosacea as a topical cosmetic seems medically reasonable.

“…Although the term “neurogenic dysregulation” has been developed in relation to rosacea in the 20 th century, the pathogenesis mechanism has been difficult to elucidate, possibly due to limitations in technologies or the heterogeneity of the disease presentation. It has been found that some neurogenic events are regarded as vital components in patients with rosacea ( e.g ., sympathetic/axon reflex-mediated alterations [ 67 ], the neuropeptide calcitonin gene-related peptide (CGRP-α) [ 68 ], substance P [ 69 ], transient receptor potential vanilloid (TRPV) [ 70 ] and the vascular endothelial-derived growth factor (VEGF) family [ 71 ]). The detection of TRP channel activation is related to inflammatory skin conditions, itching, and pain [ 25 ].…”

“…Some components of cosmetic creams used in the treatment of rosacea inhibit KLK5, such as dextran sulfate, 4‐ t ‐butylcyclohexanol, pongamia oil, and hesperidin methyl chalcone [139]. Dermasence gel increases KLK5 and suppresses CAMP in a 3D human reconstructed epidermis [140]. Cream with the protease inhibitor ε‐aminocaproic acid (ACA) improves rosacea in patients (NCT01398280) [141].…”

“…NHEKs at 70% confluence were exposed to different concentrations of recombinant human IL-33 (100 ng/ml; MACS, Auburn, CA, USA) and LL-37 peptides (0.5 μg/ml; ANYGEN, Jangseong, Korea). The expression of VEGF and TRPV1 was evaluated at times ranging from 0 to 48 h and doses ranging from 0.1 to 10 ng/ml 19 . Regulation of VEGF and TRPV1 was also evaluated after the cells were treated with citron essential oils at various concentrations (0.005%, 0.01%, and 0.02%).…”

“…Interleukin 33 (IL-33), a pro-inflammatory cytokine and an inducer of Th2-mediated responses, has been reported to enhance VEGF expression in keratinocytes 18 . In addition, abnormally increased expression of LL-37 may induce VEGF expression in epidermal keratinocytes, which corresponds to the enhanced inflammation and vascular response seen in rosacea 19 .…”
