Design and Characterization of a Silk-Fibroin-Based Drug Delivery Platform Using Naproxen as a Model Drug

Abstract: The objective of this proof-of-concept study was to develop a platform for controlled drug delivery based on silk fibroin (SF) and to explore the feasibility of using SF in oral drug delivery. The SF-containing matrixes were prepared via spray-drying and film casting, and the release profile of the model drug naproxen sodium was evaluated. Attenuated total reflectance Fourier transform infrared spectroscopy (FTIR) has been used to observe conformational changes in SF- and drug-containing compositions. SF-based… Show more

“…The silk fibroin was used as material for tissue engineering for skin, bone, blood vessel, ligament and nerve tissue regeneration, because it is non-immunogenic and non-toxic material. Some research articles show that silk fibroin is biodegradable, but the degradation rate of silk fibroin is not yet clear [163,[173][174][175][176][177][178][179][180][181][182]. The silk production is a natural ability of some arachnids, such as spiders, including some fossil species.…”

“…Once purified, fibroin can be dissolved in a number of aqueous, chaotropic high concentration salts (e.g., ≥9 M LiBr, ≥50°C or ≥10 M LiSCN, ≥25°C) or ternary systems containing alcohols (e.g., CaCl 2 –water-ethanol, ≥70°C). Subsequent desalting of high-salt fibroin solutions is generally carried out by dialysis to obtain an aqueous regenerated fibroin solution, though chromatographic desalting protocols have also been proposed [70]. …”

“…27 Preparation of sF solution SF solution was prepared as previously described. 28,29 Briefly, Bombyx mori silkworm cocoons were cut into small pieces and boiled for 30 minutes in a solution of 0.5 wt% Na 2 CO 3 . After three rinses with ultrapure water, the extracted fibroin fibers were then dried at 60°C for 6 hours.…”
