Current biosensors have limited application in clinical diagnostics as they lack the high order of specificity needed to detect low molecular analytes, especially in complex fluids (such as blood, urine, and saliva). In contrast, they are resistant to the suppression of non-specific binding. Hyperbolic metamaterials (HMMs) offer highly sought- after label-free detection and quantification techniques to circumvent sensitivity issues as low as 105 M concentration in angular sensitivity. This review discusses design strategies in detail and compares nuances in conventional plasmonic techniques to create susceptible miniaturized point-of-care devices. A substantial portion of the review is devoted to developing low optical loss reconfigurable HMM devices for active cancer bioassay platforms. A future perspective of HMM-based biosensors for cancer biomarker detection is provided.

In this study, a two-port network-based microwave sensor for liquid characterization is presented. The suggested sensor is built as a miniature microwave resonator using the third iteration of Hilbert’s fractal architecture. The suggested structure is used with the T-resonator to raise the sensor quality factor. The suggested sensor is printed on a FR4 substrate and has a footprint of 40×60×1.6mm3. Analytically, a theoretical investigation is made to clarify how the suggested sensor might function. The suggested sensor is created and put to the test in an experiment. Later, two pans to contain the urine Sample Under Test (SUT) are printed on the sensor. Before loading the SUT, it is discovered that the suggested structure’s frequency resonance is 0.46 GHz. An 18 MHz frequency shift is added to the initial resonance after the pans are printed. They monitor the S-parameters in terms of S12 regarding the change in water content in the urine samples, allowing for the sensing component to be completed. As a result, 10 different samples with varying urine percentages are added to the suggested sensor to evaluate its ability to detect the presence of urine. Finally, it is discovered that the suggested process’ measurements and corresponding simulated outcomes agreed quite well.

Better sensitivity of a biosensor could boost up the detection limit of analytes, thus a must in the fields of bio-sensing and bio-detection. To further enhance the sensitivity of a biosensor, in this work, we design an oblique-flat-sheet metamaterial perfect absorber (MPA) to concentrate the hot spots within air between the oblique flat sheet and the continuous ground metal, thus enabling fully interaction between analytes and hot spots. The corresponding field distributions in simulation corroborated our assumption and its sensitivity could be up to 1049 nm/RIU. Then, we fabricated the sample by e-beam lithography process for a seed layer and simply tilting the sample during deposition to obtain oblique flat sheets. When considering the stochastic nature of the deposited multiple oblique flat sheets, we modified the metallic upper resonator of the MPA from the single oblique-flat-sheet into randomly distributed oblique-wire-bundle (OWB) and in simulation, its sensitivity is boosted up to 3319 nm/RIU. In experiments, the measured sensitivity is 1329 nm/RIU under different concentrations of glucose solutions that is four times larger than the 330 nm/RIU of the planar MPA. The higher sensitivity was attributed to that the OWB MPA could provide hot spots within air not only between OWB and grounded metal but also among wires. Moreover, the OWB could also trap and concentrate the analytes locally.
