Context Chitosan is a biocompatible polysaccharide that has been widely exploited in biomedical and drug delivery applications. Objective This study explores the renoprotective effect of chitosan nanoparticles in vivo in rats. Materials and methods Chitosan nanoparticles were prepared via ionotropic gelation method, and several in vitro characterizations were performed, including measurements of particle size, zeta potential, polydispersity index, Fourier transform-infrared spectroscopy, differential scanning calorimetry, and transmission electron microscopy (TEM) imaging. Wistar rats were divided randomly into four groups; negative control, CCl 4 -induced nephrotoxicity (untreated), and two groups receiving CCl 4 + chitosan NPs (10 and 20 mg/kg) orally for 2 weeks. The renoprotective effect was assessed by measuring oxidative, apoptotic, and inflammatory biomarkers, and via histopathological and immunohistochemical examinations for the visualization of NF-κB and COX-2 in renal tissues. Results Monodisperse spherical nanosized (56 nm) particles were successfully prepared as evidenced by dynamic light scattering and TEM. Oral administration of chitosan nanoparticles (10 and 20 mg/kg) concurrently with CCl 4 for 2 weeks resulted in 13.6% and 21.5% reduction in serum creatinine and increase in the level of depleted reduced glutathione (23.1% and 31.8%), respectively, when compared with the positive control group. Chitosan nanoparticles (20 mg/kg) revealed a significant ( p ˂ 0.05) decrease in malondialdehyde levels (30.6%), tumour necrosis factor-α (33.6%), interleukin-1β (31.1%), and caspase-3 (36.6%). Conclusions Chitosan nanoparticles afforded significant protection and amelioration against CCl 4 -induced nephrotoxicity. Thus, chitosan nanoparticles could afford a potential nanotherapeutic system for the management of nephrotoxicity which allows for broadening their role in biomedical delivery applications.

This study aimed to create and describe mucoadhesive nimodipine solid lipid nanoparticles as buccal tablets by altering the amounts of three polymers: Carbopol 934, Hydroxypropyl methylcellulose and Hydroxyethyl cellulose. The Nimodipine-loaded solid lipid nanoparticles (SLN) were formulated by high shear homogenization and ultrasonication process using palmitic and stearic acid as the lipid matrix and Tween-80 as the surfactant. The swelling properties of all formulations were investigated, and it was discovered that all formulations have a good swelling index at 6 hours. The surface pH of each batch varied between 5.6 and 6.1. The mucoadhesive strengths (15.3-29.5 g) varied with polymer concentrations, particularly Carbopol 934. All batches had considerably different dissolution profiles, ranging from a maximum release of 89.08% (at 8h in batch NT3) to a minimum release of 80.32% (at 8h in batch NT2). SLN formulations had the best results in both Entrapment efficiency and In-vitro drug release, showing that SLN may be a promising delivery strategy for improving Nimodipine release.

In the present study, two different microneedle devices were produced using digital light processing (DLP). These devices hold promise as drug delivery systems to the buccal tissue as they increase the permeability of actives with molecular weights between 600 and 4000 Da. The attached reservoirs were designed and printed along with the arrays as a whole device. Light microscopy was used to quality control the printability of the designs, confirming that the actual dimensions are in agreement with the digital design. Non-destructive volume imaging by means of microfocus computed tomography was employed for dimensional and defect characterization of the DLP-printed devices, demonstrating the actual volumes of the reservoirs and the malformations that occurred during printing. The penetration test and finite element analysis showed that the maximum stress experienced by the needles during the insertion process (10 N) was below their ultimate compressive strength (240–310 N). Permeation studies showed the increased permeability of three model drugs when delivered with the MN devices. Size-exclusion chromatography validated the stability of all the actives throughout the permeability tests. The safety of these printed devices for buccal administration was confirmed by histological evaluation and cell viability studies using the TR146 cell line, which indicated no toxic effects.
