A new series of benzimidazole containing Schiff base moiety has been synthesized. The reaction was achieved through cyclocondensation reaction of the substituted 1,2-phenylenediamines and amino acids (glycine, alanine and tyrosine) to gave substituted benzimidazole derivatives 1(a-f). A copper-catalyzed aerobic oxidative reaction between aromatics or heteroaromatics with KSCN is developed by using O as the oxidant. The combination of Cu(OTf), N,N,N',N'-tetramethylethylenediamine (TMEDA) and BF·EtO provides an efficient catalytic system, affording substituted thiocyanation products and 2-aminobenzothiazoles in excellent yields. The reaction also possesses a good functional group tolerance for both strong electron-withdrawing and electron-donating groups.

Aerobic Copper-Mediated Domino Three-Component Approach to 2-Aminobenzothiazole Derivatives

An unprecedented three-component reaction involving a 2,2'-diaminodiaryl disulfide, copper cyanide, and an electrophile is described. This transformation is based on an oxidative copper-mediated S-cyanation as a key step and involves a cyanation/cyclization/acylation domino sequence enabling a rapid and efficient synthesis of diversely substituted 2-aminobenzothiazole derivatives. Notably, this reaction proceeds via an original mechanism involving an intermolecular migration of the acyl group.
