Using mycorrhizal fungi with their helper bacteria (MHB) to alleviate phosphate deficiency improve plant growth and rejuvenate the soil is the newer safer and most promising environment-friendly approach to limiting synthetic agrochemicals. Here 65 MHBs were isolated from the mycorrhizosphere of the Banana plant and 12 were screened based on their biofilm formation protease and N-acyl homoserine lactones production. A newly reported MHBEnterobactersp. was identified through the 16S ribotyping method and posed plant growth-promoting properties i.e solubilized phosphate produced indole acetic acid ammonia and hydrogen sulfide. The biocompatibility experiment showed a 4-fold increased mycorrhizal proliferation in MHB added plate compared to the control. Further field experiments suggested the banana plants 10-4-fold increment in plant height (216.54 cm) and stem diameter (10.66 cm). At the same time10-fold significant improvements were noted in leaf carbohydrate (639.9 mg g-1) protein (432.76 mg g-1) chlorophyll (268.66 mg g-1) phenol (1.9 mg g-1) and proline (uMole g-1) content of experimental plant compared to control. The study also showed a 5-fold increase in phosphate content (12.84 mg g-1) than the control (2.4 mg g-1). The isolates increased the mycorrhizal colonization and spore number by 79.33 % and 14.31 g-1in the rhizospheric soil. Total organic carbon and nitrogen (0.68 %) total phosphorus (55 kg ha-1) and potassium (499.66 kg ha-1) content of soil were positively affected by MHB and AMF. Further-more principal component analysis (PCA) and Pearsons correlation analysis of all the obtained results clearly showed the positive insights of inoculated MHB and AMF on the growth of the banana plant and soil restoration.

To investigate the mechanism of aquatic pathogens in quorum sensing (QS) and decode the signal transmission of aquatic Gram-negative pathogens, this paper proposes a novel method for the intelligent matching identification of eight quorum signaling molecules (N-acyl-homoserine lactones, AHLs) with similar molecular structures, using terahertz (THz) spectroscopy combined with molecular dynamics simulation and spectral similarity calculation. The THz fingerprint absorption spectral peaks of the eight AHLs were identified, attributed, and resolved using the density functional theory (DFT) for molecular dynamics simulation. To reduce the computational complexity of matching recognition, spectra with high peak matching values with the target were preliminarily selected, based on the peak position features of AHL samples. A comprehensive similarity calculation (CSC) method using a weighted improved Jaccard similarity algorithm (IJS) and discrete Fréchet distance algorithm (DFD) is proposed to calculate the similarity between the selected spectra and the targets, as well as to return the matching result with the highest accuracy. The results show that all AHL molecular types can be correctly identified, and the average quantization accuracy of CSC is 98.48%. This study provides a theoretical and data-supported foundation for the identification of AHLs, based on THz spectroscopy, and offers a new method for the high-throughput and automatic identification of AHLs.

Members of the genus Mesorhizobium, which are core components of the rhizosphere and specific symbionts of legume plants, possess genes for acyl-homoserine lactone (AHL) quorum sensing (QS). Here we show Mesorhizobium japonicum MAFF 303099 (formerly M. loti ) synthesizes and responds to N -[(2 E , 4 E )-2,4-dodecadienoyl] homoserine lactone (2 E, 4 E- C 12:2 -HSL). We show that the 2 E, 4 E- C 12:2 -HSL QS circuit involves one of four luxR-luxI -type genes found in the sequenced genome of MAFF 303099. We refer to this circuit, which appears to be conserved among Mesorhizobium species, as R1-I1. We show that two other Mesorhizobium strains also produce 2 E, 4 E- C 12:2 -HSL. The 2 E, 4 E- C 12:2 -HSL is unique among known AHLs in its arrangement of two trans double bonds. The R1 response to 2 E, 4 E- C 12:2 -HSL is extremely selective in comparison with other LuxR homologs, and the trans double bonds appear critical for R1 signal recognition. Most well-studied LuxI-like proteins use S -adenosylmethionine and an acyl-acyl carrier protein as substrates for synthesis of AHLs. Others that form a subgroup of LuxI-type proteins use acyl-coenzyme A substrates rather than acyl-acyl carrier proteins. I1 clusters with the acyl-coenzyme A-type AHL synthases. We show that a gene linked to the I1 AHL synthase is involved in the production of the QS signal. The discovery of the unique I1 product enforces the view that further study of acyl-coenzyme A-dependent LuxI homologs will expand our knowledge of AHL diversity. The involvement of an additional enzyme in AHL generation leads us to consider this system a three-component QS circuit. IMPORTANCE We report a Mesorhizobium japonicum quorum sensing (QS) system involving a novel acyl-homoserine lactone (AHL) signal. This system is known to be involved in root nodule symbiosis with host plants. The chemistry of the newly described QS signal indicated that there may be a dedicated cellular enzyme involved in its synthesis in addition to the types known for production of other AHLs. Indeed, we report that an additional gene is required for synthesis of the unique signal, and we propose that this is a three-component QS circuit as opposed to the canonical two-component AHL QS circuits. The signaling system is exquisitely selective. The selectivity may be important when this species resides in the complex microbial communities around host plants and may make this system useful in various synthetic biology applications of QS circuits.
