Detection, Structural Elucidation, and Biological Effects of Diverse N-Acyl-homoserine Lactone Signaling Molecules in the Plant-Promoting Endophytic Bacterium Rhizobium oryzihabitans M15

Abstract: Quorum sensing (QS), usually performed by N-acyl-homoserine lactones (AHLs) in Gram-staining-negative bacteria, plays an important role in plant–bacteria interactions. Rhizobium oryzihabitans M15 is a plant-growth-promoting rhizobacterium (PGPR) isolated from rice roots. In this study, we found a QS system in the endogenous plasmid of R. oryzihabitans M15 and detected the activity of AHLs by a bioassay method. We identified five AHL analogues in R. oryzihabitans M15 using liquid chromatography-tandem mass spec… Show more

“…Biofilm formation was performed using crystal violet as described by Zhao et al (2021) with minor modification (Zhao et al, 2021). Briefly, exponential phased bacterial cells (A 600 ≅ 0.05) were inoculated into a 96-well plate and incubated at 30 °C without shaking.…”

“…Isolates were spot inoculated (A 600 ≅ 0.5) at the center of the plates and incubated at 30°C. Swimming motility and zone of hydrolysis were measured, as mentioned by (Zhao et al, 2021).…”

“…A homoserine lactone ring that is linked to an acyl group is a shared characteristic of AHLs [11]. However, the acyl chain length (n = 4~18) and the substituent (hydrogen, hydroxyl, or carbonyl) on the third carbon atom exhibit diversity, which is influenced by variations in the bacterial community and environmental conditions [12]. The regulation of the expression of numerous different virulence factors by diverse AHL signaling molecules presents challenges in preventing and controlling bacterial diseases in aquaculture.…”

“…In many bacteria, AHL QS is important for success in mutualist or pathogenic interactions with eukaryotic hosts ( 1 - 5 ). The list of such bacteria includes nitrogen-fixing symbionts of legume plants, including species of Rhizobium, Bradyrhizobium, Sinorhizobium, and Mesorhizobium ( 6 - 12 ). There are a few reports on QS in the genus Mesorhizobium ( 9 , 13 , 14 ), including two, which provide evidence that QS is involved in nodulation of host legume roots ( 11 , 15 ).…”
