Development and Evaluation of a Western Blot Kit for Diagnosis of Human Trichinellosis

Abstract: We evaluated industrially prepared Western blot strips designed to avoid the cross-reactions observed with indirect immunofluorescence and enzyme-linked immunosorbent assays used for the serodiagnosis of trichinellosis. The antigen preparations were crude extracts of Trichinella spiralis. The Western blot profile characteristic of trichinellosis was characterized by comparing 60 sera from patients infected by Trichinella to 11 sera from healthy subjects, 51 sera from patients with other proven parasitic diseas… Show more

“…A muscle biopsy and/or detection of antibodies may confirm the diagnosis, but a biopsy is not mandatory. Antibodies are detectable two-three weeks after the infection, may be measured by enzyme-immune assay, immunofluorescent, indirect hemagglutination, precipitin and bentonit flocculation assays, and a western-blott kit is under development (Capo, 1996;Grove, 2000;Yera et al, 2003). Trichinosis is very rare in Turkey.…”

“…A reduction of 75.5 % in the parasitic load was observed, when the mice infected with 10 T. spiralis muscle larvae were treated by using under-dosed with albendazole (20 mg/kg for 7 days) at 40 dpi (de-la-Rosa et al, 2007). While albendazole (200 mg/kg for 7 days) was given to the mice infected with 100 larvae at 35 dpi, the larval reduction rate was 71.8 % at 15 days after treatment (Xue et al, 2010). The efficacy of albendazole against the larvae in muscle tissues depend on the time between infection and treatment and could be dose-dependent.…”

“…different bands have been reported as Trichinella-specifi c by different authors. For example, Yera et al [35] claimed T. spiralis antigenic components with Mw of 43-44 and 64 kDa as discriminative for Trichinella infection. In a study performed by Gomes-Morales and colleagues [36] another threeband pattern, ranging in size from 53-72 kDa (for human sera) and 48-72 kDa (for pig sera) was chosen for it discriminative characteristics.…”
