Development and linkage mapping of E-STS and RGA markers for functional gene homologues in apple

Abstract: Linkage maps developed from known-function genes can be valuable in the candidate gene mapping approach. A set of 121 expressed sequence tagged site (E-STS) primer pairs were tested on a framework genetic linkage map of apple (Malus x domestica Borkh.) constructed using simple sequence repeats (SSRs) and randomly amplified polymorphic DNA (RAPD) markers. These known-function gene markers, E-STSs, were supplemented by markers for resistance gene analogues (RGAs), designed based on conserved motifs in all charac… Show more

“…The apple molecular map currently has more than 900 markers on 17 linkage groups (Maliepaard et al, 1998;Liebhard et al, 2002Liebhard et al, , 2003aLiebhard et al, , 2003bXu and Korban, 2002b;Naik et al, 2006). Several efforts are under way to develop a substantial resource of molecular markers for genotyping and marker-assisted selection (MAS; Tartarini et al, 2000;Liebhard et al, 2002Liebhard et al, , 2003aLiebhard et al, , 2003bSilfverberg-Dilworth et al, 2006).…”

“…Several sequencing projects have generated more than 250,000 expressed sequence tags (ESTs) in apple over the past few years (Newcomb et al 2006, Naik et al 2006. SSRs associated with these sequences constitute a large resource of markers which have remained largely unexploited.…”

“…The percentage resistant seedlings in these two families changed to 55 and 88%, respectively, in 2004, and 32 and 37% in 2005, rather than the 50% one would expect for a dominant gene in heterozygous form in Antonovka. The major-gene hypothesis was also not supported by mapping studies of the Antonovka debnicka )Summerred population generated earlier (Quamme et al 2003;Naik et al 2006). Although the nature of resistance in these two Antonovka variants remains unclear, it was consistently transmitted to a high proportion of their progeny ( Fig.…”
