Topical curcumin shows poor local availability because of its low aqueous solubility and inadequate tissue absorption. Curcumin nanocrystals were prepared by sonoprecipitation followed by lyophilization to improve surface area and solubility. The formulation was optimized by the Design of Experiment (DoE) approach. The nanocrystals were characterized for particle size, zeta potential, polydispersity index, scanning electron microscopy (SEM), powder x-ray diffraction (PXRD), practical yield and in vitro drug release studies. The nanocrystal-incorporated gel was evaluated for drug content, ex vivo permeation, in vivo skin irritation, and in vivo wound healing activity. Time of sonication and amplitude influenced the optimization of curcumin nanocrystals, but the effect of stabilizer concentrations was not significant beyond 0.5% w/w. SEM images of curcumin nanocrystals revealed irregular and plate-shaped particles with rough surfaces. PXRD patterns of curcumin nanocrystals showed low crystallinity compared to unprocessed curcumin powder. An in vitro drug release study demonstrated significant improvement in the percentage cumulative drug release in the form of nanocrystals compared to the unprocessed curcumin, and the release profile exhibited first-order kinetics. Curcumin nanocrystal gel showed 93.86% drug content and was free of skin irritation potential. Excision wound healing activity in albino rats showed that the curcumin nanocrystal gel exhibited significantly faster wound contraction than curcumin powder-incorporated gel.

Patches are punched from a sheet that serves as the intermediate product. Oral mucosal medication delivery alternatives to tablets have been studied, including polymers like cellulose etc. can be used to create mucosal patches. The present research was based on the development and in-vitro characterization of mucoadhesive patches of root extract of Adenium obesum utilizing different polymers. Adenium obesum (fresh roots) was obtained from the Unnao region. Eudragit L-100, Propylene glycol, Tween 80, Methanol, Ethanol and HPMCK 4M & HPMCK 15M were purchased from the local market. The initial stages of processing involve grinding the roots into a coarse powder. The Soxhlet equipment was used to extract the powder and then weigh it. After preformulation study, the Adenium obesum mucoadhesive patches were created using the solvent casting technique. Patches were evaluated for physical appearances, thickness, pH, % drug content, in-vitro drug release, SEM and stability. In results, an excellent % drug release was demonstrated by the buccal patch. In order to estimate stability profile of mucoadhesive patches, it was evaluated for physical appearance. In conclusion, patches i.e., F2, F3 and F6 exhibited a significant estimation parameters like drug content, % drug release etc. The buccal drug of herbal (A. obesum) mucoadhesive patch has shown improved stability. It will ease to provide the long-lasting action by suppressing the symptoms of insomnia, agitations etc. Therefore, it may be produced commercially after its successful clinical trials.

Purpose:The long term oral therapy of vildagliptin is associated with potential hepatotoxicity and low patient compliance. This study aims at development of a drug in adhesive transdermal patch system of vildagliptin using pressure sensitive acrylic polymers and compare the pharmacokinetics with oral therapy. Methods: The prospective of different chemical enhancers in improving the transport of vildagliptin was assessed ex vivo. In vivo permeation studies in rats were performed using patch system contain sodium lauryl sulfate as enhancer. Results: Drug solubility varied among adhesive bases tested and maximum value (15% w/w) observed with Duro-Tak 87-2510. Incorporation of chemical agents significantly improved the permeation of vildagliptin, but not to the same extent. The highest flux value of vildagliptin was obtained by the addition of sodium lauryl sulfate (22.96 ± 3.58 µg/cm 2 /h; P<0.0001), which is ~4 folds higher as compared to control. Pharmacokinetic profiles of vildagliptin were different for transdermal and oral delivery with significantly high bioavailability by transdermal delivery. The AUC 0-α in transdermal delivery (1018.43 ± 79.56 ng.h/mL) was ~14 folds higher (P<0.0001) as compared to oral administration. Conclusion: The current study concludes that the fabricated drug in adhesive transdermal system could be employed for the effective delivery of vildagliptin.
