Development and validation of an LC‐MS/MS method for the determination of mesalazine in beagle dog plasma and its application to a pharmacokinetic study

Abstract: A simple, specific and sensitive LC-MS/MS method was developed and validated for the determination of mesalazine in beagle dog plasma. The plasma samples were prepared by protein precipitation, then the separation of the analyte was achieved on a Waters Spherisorb C6 column (150 × 4.6 mm, 5 µm) with a mobile phase consisting of 0.2% formic acid in water-methanol (20:80, v/v). The flow rate was set at 1.0 mL/min with a split ratio of 3:2. Mass spectrometric detection was achieved by a triple-quadrupole mass spe… Show more

“…Mesalazine chemical structure (7) MES is an antioxidant and derivate of salicylic acid that traps free radicals, which are potentially harmful species by-products of metabolism. It is a major metabolite present in blood with a half-life of up to 10 h (8) With its significance in the pharmaceuticals industry, quality control in pharmacopeia through various analytical methods has been used to analyze MSA in actual samples, such as liquid chromatography (LC) (9) With its significance in the pharmaceuticals industry, quality control in pharmacopeia through various analytical methods has been used to analyze MES in actual samples, such as chromatographic (10,11,12,13) . There are other methods which depend on the reaction of Schiff bases forming (14) and also reactions of forming charge transfer complexes (15) and chromatographic methods such as (UPLC) (17) .Among these, electrochemical methods were found to be useful for rapid response, sensitive and selective determination of various pharmaceutical applications (16,17) .…”

“…Various analytical methods were described in the literature for the determination of 5-ASA, such as liquid chromatography coupled to mass spectrometry for drug quantification in plasma samples [3], HPLC with UV detection for stability-indicating assays [4] and analysis in pharmaceutical formulations [5], capillary electrophoresis for drug impurity profiling [6], spectrophotometry [7], fluorescence spectroscopy [8], automated chemiluminescence [9] and ultra-performance liquid chromatography [10]. At present, several HPLC methods with a fluorescence [11][12][13], electrochemical [14,15] and mass spectrometry detection [16,17] were developed for simultaneous quantification of 5-ASA and its N-acetylated metabolite.…”

“…So, decorated release agents are designed for drug delivery at the terminal ileum and colon. 5 At present, the generally recognized methods for studying the structure and performance of 5-ASA include spectrophotometry, 6,7 high-performance liquid chromatographic, 8 electronic chemistry 3 and liquid chromatography-mass spectrometry, 9,10 etc. However, there is a little means for the detection of 5-ASA in drug and biological samples, due to the complexity of 5-ASA derivatization process, cumbersome extraction process and expensive instruments, and low detection capacity.…”
