Development and validation of an HPLC-DAD and HPLC/ESI-MS2 method for the determination of polyphenols in monofloral honeys from Tuscany (Italy)

Objective: The Anadenanthera colubrina (Vell.) Brennan var. cebil is a medicinal plant that has been used for the treatment of many diseases in the northeastern region of Brazil. This plant contains secondary metabolites such as quercetin, a flavonoid that is known by its antioxidant and antiinflammatory effects. The aim of this work is to propose the validation of an analytical method using high-performance liquid chromatography with diode array detector (HPLC-DAD) for the quantification of quercetin and standardization of the hydroalcoholic extract (HAE) of A. colubrina. Methods:The A. colubrina extracts were prepared by the maceration process with powdered leaves at 20% weight: volume (w/v) and a hydroalcoholic solution at 50% volume: volume (v/v) for 120 h at room temperature. After pretreatment of the hydroalcoholic extract, the quercetin marker was used for quantification and proceeded to the evaluation of validation parameters for the method using HPLC-DAD. Results:The analytical method proved to be specific. Linear over the range 1.4-26.6 µg/ml, regression analysis showed a good correlation coefficient (R2= 0.999); the limit of detection (LOD) and the limit of quantification (LOQ) were 0.27 and 0.81 μg/ml respectively. The relative standard deviation (RSD) did not exceed 2.5% for precision. The proposed method was validated with an average recovery of 92.5-97.5%. Conclusion:The method was validated using HPLC-DAD, allowing the quantification of quercetin in the standardisation process of extracts and quality control of the herbal drug containing A. colubrina Phyto complex.

Dietary intervention with Stevia residue extracts alleviates impaired glucose regulation in mice

The objective of this study was to investigate the effect of Stevia residue extracts (SREs, i.e., SRE‐1 and SRE‐2) on impaired glucose regulation (IGR). ICR mice were fed with a high‐fat and high‐fructose diet (HFFD) to induce IGR. Glucose metabolism, serum lipid profiles, serum antioxidant status, and hepatic histopathological analysis by oil red O were analyzed. The bioactive compounds of SREs were identified by LC/MS/MS. Chemical analysis of SREs revealed the extracts to be good sources of chlorogenic acids and flavonoids. SRE‐1 significantly lowered blood glucose levels and improved glucose intolerance in HFFD‐induced IGR mice. Serum lipid profiles, antioxidant status, and liver fat accumulation were also improved by SRE‐1, especially at 200 mg/kg. The results suggest that SRE‐1‐enriched diet could ameliorate prediabetic associated metabolic disturbances by regulating glucose and lipid pathways. Moreover, SRE‐1 exerts its beneficial effect against IGR probably due to the presence of chlorogenic acids and flavonoids. Practical applications Stevia rebaudiana Bertoni, an ancient perennial shrub of South America, produces steviol glycosides that are low calorie sweeteners, about 300 times sweeter than saccharose. In addition to steviol glycosides, S. rebaudiana contains other metabolites with bioactive potential, such as polyphenols. A large amount of byproduct is being formed during steviol glycosides extraction. Thus, there has been an increasing demand to exploit this polyphenol‐rich byproduct as food additives or nutraceuticals. Our study showed that SREs‐enriched diet has a variety of beneficial effects on HFFD‐induced IGR mice by regulating oxidative stress, glucose, and lipid metabolism. IGR refers to a metabolic status between normal glucose homeostasis and diabetes, and is associated with an increased risk of type 2 diabetes and cardiovascular disease. Therefore, SREs could be developed as functional foods or nutraceuticals to prevent many diseases associated with IGR.

Honey as a Sole Treatment of Type 2 Diabetes Mellitus

Objectives: The complications from type 2 diabetes mellitus (DM) still occur despite the ongoing efforts to prevent, or decrease it. This clinical trial aimed to evaluate the effects of honey, as an alternative, non-expensive natural substance in twenty adult patient volunteers suffering from type 2 DM and its associated metabolic derangements. Methods:The patients volunteered to stop the medicines and use honey as a sole treatment of their illnesses. There was no specific dietary restriction except for food preservatives and additives, beverages and sweets.Results: Honey consumption resulted in more hyperglycemia in these patients, but without diabetic ketoacidosis (DKA) or hyperglycemic hyperosmolar state (HHS). Longer-term honey consumption resulted also in weight reduction in all the patients, and control of the blood pressure in the patients, who had hypertension before the honey intervention. The median duration of the honey intervention, without anti-diabetic medicines was 1.09 years, with a range from 0.42 to 13.5 years. Ten of the 20 patients continued the honey intervention for more than 1 year, whereas the other 10 continued the intervention for one year or less. The only cause of the discontinuation of the honey intervention was persistent hyperglycemia. Despite persistent hyperglycemia in all the patients, and persistence of the dyslipidemia in the patients, who had dyslipidemia before the intervention, the macro-vascular complications, particularly the coronary heart disease (CHD), did not develop in any of them. On the contrary, the cardiovascular status improved in the patients, who had CHD before the intervention. However, micro-vascular complications developed in two patient volunteers, who continued the honey intervention, without medicines, for more than 8 years. Conclusion:The author imposed on announcing these results, which might be of help for the patients with diabetes mellitus.
