This study focuses on the development of a nanosupport based on halloysite nanotubes (HNTs), Fe3O4 nanoparticles (NPs), and thiolated chitosan (CTs) for laccase immobilization. First, HNTs were modified with Fe3O4 NPs (HNTs-Fe3O4) by the coprecipitation method. Then, the HNTs-Fe3O4 surface was tuned with the CTs (HNTs-Fe3O4-CTs) by a simple refluxing method. Finally, the HNTs- Fe3O4-CTs surface was thiolated (-SH) (denoted as; HNTs- Fe3O4-CTs-SH) by using the reactive NHS-ester reaction. The thiol-modified HNTs (HNTs- Fe3O4-CTs-SH) were characterized by FE-SEM, HR-TEM, XPS, XRD, FT-IR, and VSM analyses. The HNTs-Fe3O4-CTs-SH was applied for the laccase immobilization. It gave excellent immobilization of laccase with 100% activity recovery and 144 mg/g laccase loading capacity. The immobilized laccase on HNTs-Fe3O4-CTs-SH (HNTs-Fe3O4-CTs-S-S-Laccase) exhibited enhanced biocatalytic performance with improved thermal, storage, and pH stabilities. HNTs-Fe3O4-CTs-S-S-Laccase gave outstanding repeated cycle capability, at the end of the 15th cycle, it kept 61% of the laccase activity. Furthermore, HNTs-Fe3O4-CTs-S-S-Laccase was applied for redox-mediated removal of textile dye DR80 and pharmaceutical compound ampicillin. The obtained result marked the potential of the HNTs-Fe3O4-CTs-S-S-Laccase for the removal of hazardous pollutants. This nanosupport is based on clay mineral HNTs, made from low-cost biopolymer CTs, super-magnetic in nature, and can be applied in laccase-based decontamination of environmental pollutants. This study also gave excellent material HNTs-Fe3O4-CTs-SH for other enzyme immobilization processes.

Development and validation of liquid chromatography method for simultaneous determination of multiclass seven antibiotic residues in chicken tissues

Background Antibiotics are routinely used on poultry for therapy and prevention of diseases and to enhance animal growth. The objective of this study was to develop and validate a sensitive and reliable liquid chromatography with UV detection (LC-UV) method for the simultaneous determination of seven multiclass antibiotic residues (amoxicillin, ampicillin, penicillin, sulfamethoxazole, gentamicin, ciprofloxacin, and erythromycin) in chicken tissues. Methods The liquid chromatography method with UV detection was optimized for complete separation of the seven selected antibiotic compounds with reversed phase and isocratic elution using Hypersil BDS-C18 (3 µm, 100 mm × 4 mm) column. The mobile phase consisted a ratio of 0.05 M Na2HPO4, acetonitrile and methanol (70:10:20), at UV absorption wavelength of 230 nm. The column thermostat was set at 40 °C, the mobile phase flow rate was 1 mL min−1, and the injection volume was 20 μL. Results All the seven standard compounds were eluted within 14 min. The results for: linearity, precision, sensitivity, accuracy, specificity, decision limit (CCα), detection capability (CCβ), suitability and method robustness were validated according to the criteria of Commission Decision 2002/657/EC guidelines. Calibration plot correlation coefficients ranged from 0.9983 to 0.9998 and the percent relative standard deviations for repeated analysis were below 5% indicating acceptable method precision. The limits of detection (LODs) and quantification (LOQs) ranged from 0.098–0.255 μg kg−1 to 0.297–0.574 μg kg−1, respectively. The accuracy study yielded recoveries in the ranges 98.1–107% for the pure compounds and 94.0–102% for the spiked drug free chicken tissue samples. Conclusions The method was found to be appropriate for simultaneous determination of five different classes of seven antibiotic residues in chicken tissues. Furthermore, this is the first instance for the simultaneous determination of seven multiclass, multi-residues analysis using LC-UV from chicken tissue samples. This is a cost-effective and alternative method with simple instrumentation approach for laboratories that lack highly specialized state-of-the-art instrumentation.

Background Many analytical methods for testing amoxicillin trihydrate from different monographs such as United States and British pharmacopoeia, use acetonitrile HPLC grade as an organic solvent in the mobile phase; however, this solvent is expensive and not environmentally friendly Objectives Developing and validating a simple, affordable, accurate, precise and environmentally friendly HPLC-UV method, for determining amoxicillin in formulations by using methanol HPLC grade as an organic solvent in the mobile phase Methods An HPLC system was used for developing and validation of laboratory test method which is less expensive and uses environmentally-friendly mixture of mobile phase solutions. Specificity, linearity, precision, repeatability, and accuracy were studied. Results The retention time (RT) for amoxicillin was 3.53 ± 0.020 min, and no interfering peaks were recorded with the blank, standard and sample at RT, ensuring specificity. Calibration curve of 20 to 160μg/ml was used. The correlation coefficient (r2) =0.9998, which indicates that the method has the linearity to this range of 20 to 160μg/ml. Intra- and between days' repeatability were assessed by injecting solutions three times a day and within three days. The %RSD of 0.3% and 0.7% within and between days respectively, were recorded. The %RSD was ≤ 2%, which indicates a precision of the method. An average percent recovery of 100.5±3.6% was recorded. Conclusion An environmentally-friendly, simple, affordable, selective, specific, rapid, sensitive, repeatable, with precision and accurate HPLC-UV method, has been developed and validated for the estimation of amoxicillin trihydrate in pharmaceutical dosage forms and can be adopted for the purpose of quality control.
