Development of a quantitative NS1-capture enzyme-linked immunosorbent assay for early detection of yellow fever virus infection

Abstract: Yellow fever is an arboviral disease that causes thousands of deaths every year in Africa and the Americas. However, few commercial diagnostic kits are available. Non-structural protein 1 (NS1) is an early marker of several flavivirus infections and is widely used to diagnose dengue virus (DENV) infection. Nonetheless, little is known about the dynamics of Yellow fever virus (YFV) NS1 expression and secretion, to encourage its use in diagnosis. To tackle this issue, we developed a quantitative NS1-capture ELIS… Show more

“…Although frequently used for the diagnosis of DENV in countries where it is endemic, few reports have described the specific detection of NS1 antigen as a diagnostic test for YFV (previously identified in YFV studies as gp48). Ricciardi-Jorge et al described the development of a quantitative YFV NS1-capture ELISA with 80% sensitivity (12/15 cases) and 100% specificity compared to those of RT-PCR (85). NS1 concentrations ranged from 177.6 to 4,597.6 ng/ml of serum and the 3 negative samples came from patients on days 4 (n ϭ 2) and 7.…”

“…Zika virus (ZIKV) secretes NS1 efficiently from infected cells. Other Flaviviruses like Yellow Fever virus (YFV) also secrete NS1 albeit in very low amounts compared with DENV or ZIKV [24]. In order to compare with DENV, we tested the secretion properties of ZIKV and YFV NS1 in infected C6/36 and Vero-E6 cells.…”

“…Another interesting observation is that the CBD is fully conserved in DENV and ZIKV NS1, while incomplete in other flaviviruses such as YFV, JEV and WNV. Secretion of YFV NS1 have been observed in mosquito cells albeit a higher concentration was maintained as cell-associated rather than secreted into the extracellular milieu (28); moreover, no NS1 secretion is observed in WNV or JEV infected mosquito cells (29, 30). Thus, the presence of an active CBD and the recognition of NS1 by the mosquito caveolin may be crucial for NS1 secretion in the mosquito, and suggest unique roles for the DENV and ZIKV soluble NS1 in the mosquito (11, 21).…”
