Development of a Radioimmunoassay for Canine Luteinizing Hormone

Abstract: A double antibody radioimmunoassay for canine luteinizing hormone (LH) was devised. The assay can measure immunoreactive LH in the peripheral plasma of the normal male and female dog. An increase in hormone concentration was found on the 2nd day of oestrus in the female. Ovariectomy raised plasma concentrations of the hormone to levels greater than those observed during oestrus. The administration of LH-releasing factor induced a rapid increase in the plasma concentrations of LH.

“…Progesterone rose late in oestrus, reached peak values during the first half of pregnancy and fell gradually towards term. The patterns of IRCLH and progesterone were essentially similar to those previously observed in the non-pregnant animal (Boyns et al 1972; Parkes et al, submitted for pubi.). Serial dilutions of plasma samples taken at oestrus and during pregnancy were parallel to the standard curve.…”

“…Details of the preparation and double antibody radioimmunoassay of canine luteiniz¬ ing hormone have been given elsewhere (Stockell Hartree et al 1972: Boyns et al 1972). Antiserum (first antibody) to canine lutcinizing hormone was raised in a rabbit and used at a final dilution of Vsoooo.…”

“…RIA reagents for rat PRL, dog PRL, GH and LH were kindly supplied by NIAMDD (Baltimore, MD, USA). The method of RIA for dog PRL (20), GH (21) and LH (22) were previously described. Their levels were expressed as ng/ml of serum.…”

“…Those con¬ taining the highest concentrations of prolactin were pooled, divided into 1 ml portions (containing approximately 60 fig protein) and lyophilised. This material was designated PPGE-1.Radioimmunological assay using This was performed as for canine LH(Boyns et al 1972). PPGE-1 (5 ug) in 25 fil 0.05 M phosphate buffer pH 7.5 was iodinated by a modification of the method ofGreenwood et al (1963) previously described(Boyns et al 1972).…”
