Development of a rapid resolution HPLC method for the separation and determination of 17 phenolic compounds in crude plant extracts

Abstract: Abstract:© Versita Sp. z o.o. Keywords: Plant phenolics • Rapid Resolution HPLC • Medicinal plantRapid Resolution HPLC/DAD method, on a 1.8 μm, 4.6×50 mm column, was developed to enable a rapid separation of a mixture of 17 compounds, which consisted of hydroxybenzoic acids, hydroxycinnamic acids, flavones, flavonols, flavanone, flavonol-glycoside and antraquinone, in a single run, within 22 minutes. The developed method is precise, accurate and sensitive enough for simultaneous quantitative evaluation of maj… Show more

“…The crude leaf extract of A. cathartica L. was subjected to initial separation by TLC where Benzene : Ethyl acetate (1:1) solvent systems exhibited most effective separation with three bands. However, it was our primary attempt to separate active single compound from the crude leaf extract but to identify and characterize the bioactive compound more preciously, the crude leaf extract of A. cathartica L. can be further subjected to other chromatographic techniques (Mišan et al, 2011;Sasidharan et al, 2011;Patra et al, 2012).…”

“…Selected medicinal plants and the mixture were already shown to be a rich source of plant phenolics [8] and to possess antioxidant activity by in vitro tests [7]. A relatively high in vitro antioxidant activity of "Vitalplant" mixture was explained by synergistic effects of its components [7].…”

“…The extraction of phenolic compounds from the flour was performed in accordance with the method described in Jambrec et al (2015), while the HPLC/DAD analysis was consistent with the procedure argued by Mišan et al (2011). The procedure is shown in Figure 1.…”
