Development of an improved medium for germination of Cajanus cajan (L.) Millsp. pollen in vitro

Abstract: A simple, reliable medium for pollen germination of Cajanus cajan was developed by modifying Brewbaker and Kwack (BK) medium. Past attempts of C. cajan pollen germination in artificial media were not successful. A medium containing polyethylene glycol 4000 (PEG) showed more than 90% germination for C. cajan var. Pusa 33 only when the young buds (36 h before anthesis) were kept in pollen germination medium (PGM) for 36 h before pollen extraction. Supplementation of PGM with epsilon-amino caproic acid (EACA), an… Show more

“…An alternative medium with 10% PEG (polyethylene glycol 4000) was modified from [53], [54] and contained, 0.01% H 3 BO 3 , 3 mM Ca(NO 3 ) 2 , 1 mM MgSO 4 , 1 mM KNO 3 , 10% (w/v) PEG, 10% (w/v) sucrose, pH 7.5 with KOH. To make solid medium with PEG, the liquid medium minus PEG was first solidified with 1% low-melting agarose, and then equilibrated with liquid medium including 10% PEG.…”

“…Pollen of other tropical legumes such as pigeonpea ( Cajanus cajan L.; Jayaprakash and Sarla 2001) germinates at room temperature (20 ± 1°C). Interestingly, Phaseolus , also a warm season legume, requires low temperatures for in vitro pollen germination.…”

“…It was composed of 100mg 1-1 boric acid, 200mg 1-1 magnesium sulfate, 100mg 1-1potassium nitrate, 300-mg 1-1 calcium nitrate, 1% agar and sugars (Jayaprakash, & Sarla, 2000). Five sugars were utilized, namely; fructose, glucose, lactose, maltose and sucrose.…”
