Development of molecular diagnostics for the detection of Ganoderma isolates pathogenic to oil palm.

Abstract: This chapter focuses on the development of a rapid molecular diagnostic method for the detection of Ganoderma pathogenic to oil palm through the EU-STABEX-funded programme at the Papua New Guinea Oil Palm Research Association. The internal transcribed spacer (ITS) region of oil palm-associated isolates, selection of primer sites, evaluation of primer combination and their diagnostic capabilities are described. The ITS-based approach provides a single diagnostic method for the taxon which is independent of the … Show more

“…One of these sections is particularly suited to the generation of a PCR primer. Bridge et al2000 [5], Pilotti et al 2002 [12] This primer is specific to G. boninense when used in conjunction with a universal ITS primer. DNA from environmental samples was tested and compared to the other Ganoderma species.…”

“…Determination of Ganoderma isolates using molecular approaches was proposed by Bridge et al, (2000) for Ganoderma cultures originated from environmental samples. Therefore, molecular analysis with SSU-817 and SSU-1536 primer set to target at the 18S small sub-unit rDNA region was conducted on the 12 Ganoderma isolates listed in (Idris, 1999 andMiller, 1995).…”

“…As no IAC was included, the conclusions about species are invalid, and the use as a diagnostic is based on a false premise at least. Bridge et al. (2000) is another case for Ganoderma .…”
