Abstract:In vitro multiplication of date palm is a very promising way for the rehabilitation of Moroccan palm devastated. The objective of this work was to study the development of vitroplants introduced into traditional and modern palm of the Errachidia region. The procedure followed is to make descriptive diagnoses in different sites where the palm was introduced and to evaluate the mortality of plants and cultural practices. The mortality rates varied from 78% to 19% in new plantation depending on the cultivation technique used. At the traditional palm, the rate of recovery of plantlets was zero. The mortality is higher in Nejda cultivar compared to Boufegouss. This variety also presented a better adaptation in terms of shoot height in comparison with the development of Nejda and Mejhoul varieties. The production of offshoots as well as inflorescences takes place three years after plantation. The importance of improving cultivation techniques in date palm for the development of this sector in Morocco is discussed.

The Moroccan demand for date palm plants over the next decade is estimated to be 2.9 million. To fulfill this huge demand, the use of rapid micropropagation techniques is an objective of great interest. Generally, date palm micropropagation is performed from offshoot tissue. However, in the case of some rare genotypes, this technique is inefficient because only limited numbers of offshoots are available. To overcome this problem, a new technique based on the use of emerged inflorescence tissue was developed. The research activities resulted in a completely successful process starting from plant material excision to plant acclimatization and field planting. The technique was used for the micropropagation of 16 date palm genotypes with good fruit quality. To date, hundreds of well-acclimatized plants belonging to 9 genotypes have been produced. In addition, clusters of buds of 3 selected genotypes and Medjool cv. were produced and transferred to a private laboratory for mass propagation. In vitro plants needed to test for bayoud disease resistance of 6 selected genotypes were produced using this technique. Since 2001, well-acclimatized in vitro plants were gradually transferred to the field to study their behavior. Inflorescence-derived plants have shown normal growth and no abnormalities. Fruit set of planted palms started in 2005. In the present paper, micropropagation with inflorescence tissue as well as the related major achievements are presented.

The effect of basal medium, explant size and density on shoot multiplication, growth, rooting and acclimatization of date palm cv. '16-bis' was evaluated. Bud clusters of different sizes (2, 3, 4 and 5 buds per cluster) were cultured at density of 1, 2, 3 and 4 clusters on Murashige and Skoog medium (MS), woody plant medium (WPM) and Nitsch medium (NM) supplemented with 0.5 mg/L 2-naphthoxyacetic acid and 0.5 mg/L kinetin for three months (multiplication phase). Separated shoots of different sizes (<3 cm; 3 to 4.5 cm and 4.5 to 6 cm) were cultured at density of 1, 2, 3 and 4 shoots on hormone free MS medium, WPM or NM for three months (Elongation-rooting phase). The proliferation and development of shoots were affected by the basal medium, explant size and density. The optimal shoot proliferation (18.1) was observed when 4 buds clusters were cultured at the density of 2 clusters per jar in MS medium. Separated shoots of 4.5 to 6 cm length exhibited the optimal in vitro development in terms of leaf length and greening, and root number and length when cultured on MS medium. In addition, these shoots reached the highest acclimation frequency with 80%. Our results would be utilized for an efficient propagation of plantlets of cv. '16-bis' , a selected date palm cultivar resistant to the bayoud disease.
