Proniosomes are drug-encapsulated, nanoscale vesicular structures that generate multilamellar niosomal dispersions upon hydration. This study aimed to develop enalapril maleate nanoproniosomal gels and assess their effectiveness in experimental hypertensive rat models. The gels were synthesized based on the coacervation phase separation method, using lecithin, cholesterol, and various nonionic surfactants as formulation components, along with the drug. The developed gels were then subjected to various analyses, such as pH, viscosity, rate of spontaneity, entrapment efficiency, vesicle size, ex vivo permeation, skin irritation, scanning electron microscopy, stability, in vivo bio-availability, in vivo antihypertensive activity, and in vitro-in vivo correlation studies. Results revealed that all synthesized gel formulations maintained good physical characteristics, within permissible limits. The results of the ex vivo skin permeation analysis revealed non-Fickian release kinetics and zero-order penetration behaviors of the drug formulations with diffusion, achieving a cumulative permeation rate of 58.75%-89.72% through albino rat skin over 24 h. Moreover, skin irritation tests revealed that the topical application of the drug formulations did not cause any signs of irritation, indicating their safety. Furthermore, in vivo bio-availability studies revealed that one particular formulation, EMNP7, demonstrated an approximately 188.99-fold greater bio-availability compared to the Vasotec tablet. Additionally, in vivo antihypertensive analysis revealed that this formulation effectively restored elevated rat blood pressures to the normal range. Furthermore, the in vitro-in vivo correlation analysis suggested that the ex vivo (in vitro) data could accurately replicate in vivo physiological conditions. Overall, our findings indicate that enalapril maleate encapsulated within nanoproniosomal gels can effectively function as controlled drug delivery systems, releasing the drug once per day for effective hypertension management.

Proniosomes, prepared in dry form and hydrated by agitation in hot water to form niosomes provide an alternative with prospective for drug delivery via the transdermal route. The transdermal route of drug delivery has many advantages for administration of drugs in local and systemic therapy. But skin is widely recognized for its effective barrier properties compared with other biological membranes. The low permeability of the skin makes it a dermal delivery is an alternative route, but dermal delivery is an alternative route. Newer drugs of lipophilic nature emerge poor bioavailability, irregular absorption, and pharmacokinetic changes. Therefore, this novel drug delivery system has been proved advantageous over other oral and topical delivery of drug candidates to bypass such disruption. This Proniosomal gel basically is a compact semi-solid liquid crystalline (gel) composed of non-ionic surfactants easily formed on dissolving the surfactant in a minimal amount of acceptable solvent and the least amount of aqueous phase and phosphate buffer. Topical application of gel under occlusive condition during which they are converted into nisomes due to hydration by water in the skin present itself. Proniosomal gels are typically present in transparent, translucent, or white semisolid gel texture, which makes them physically stable throughout storage and transport. This review provides an important overview of the preparation, formulation, evaluation, and application of Proniosomal gel as a drug delivery carrier.

Nanotechnology is an emerging technology that has brought the upheaval reforms in the domain of pharmaceutical sciences including the development of nanovesicular drug delivery carriers such as proniosomes, niosomes, liposomes, ethosomes, etc. Among them, proniosomes become superior and they surmount the problems of other vesicular carriers. Proniosomes are nano-sized vesicular structures of dry, free-flowing powder (or) gel with encapsulation of drugs in the vesicle that produce multilamellar niosomal dispersion after hydration and they possess the capacity to enhance solubility, permeability and bioavailability of diverse drugs. Proniosomes are suitable to deliver the drugs efficiently through transdermal route to accomplish controlled release action, increased therapeutic effectiveness to various diseases and upon application to the skin, the proniosmes are modified into niosomes in situ by hydration of water from the skin. This study aims to discuss different aspects of proniosomes such as merits, mechanism, types, components, preparation, characterization, drug release, market scenario, future trends and to explore proniosomes for various pharmaceutical applications in drug delivery via different routes, such as topical, transdermal, oral, parenteral, ocular, vaginal, nebulizer and intranasal routes. These proniosomes-derived niosomes are better and may offer an excellent, inexpensive alternative delivery, much more beneficial than other vesicular and conventional dosage forms.
