Development, validation and transfer of a hydrophilic interaction liquid chromatography/tandem mass spectrometric method for the analysis of the tobacco‐specific nitrosamine metabolite NNAL in human plasma at low picogram per milliliter concentrations

Abstract: A highly sensitive bioanalytical method based on a simple liquid/liquid extraction and hydrophilic interaction liquid chromatography with tandem mass spectrometry (HILIC/MS/MS) analysis has been developed, validated and transferred for the determination of 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL), a tobacco-specific nitrosamine metabolite. Deuterated NNAL (NNAL-d(4)) was synthesized and used as the internal standard. This method can be used for the analysis of free and total NNAL (free NNAL plus NN… Show more

“…Urinary nicotine and its metabolites were analysed using solid phase extraction and liquid chromatography with tandem mass spectrometry based on the method described by St. Charles and colleagues [26]. NNAL and its metabolites were analysed using a solid phase extraction and a LC/MS/MS method based on that of Xia and colleagues and Pan and colleagues [27,28].…”

“…Eight biomarkers of exposure were measured in 24h urine from these groups (nicotine, cotinine, hydroxycotinine, NNAL and all of the respective glucuronide conjugates). NNAL and its metabolite were analyzed using a solid phase extraction and liquid [11] and Pan et al [12] (LLOQ 5ng/ml ULOQ 1000ng/ml). Urinary nicotine and its metabolites were analyzed using a solid phase extraction and liquid chromatography with tandem mass spectrometry as described by St. Charles et al [6] (LLOQ 10ng/ml ULOQ 5000ng/ml).…”

“…Use of standardized automation equipment has shown to be advantageous during the method transfer. Unfortunately very limited information on method transfer can be found in the literatures [35,149]. Typically in our organization, before the method transfer, scientists from both sites go through the method details very carefully.…”
