Developmental aspects of polyamine-oxidizing enzyme activities in the mouse kidney. Effects of testosterone

Abstract: In the present study developmental patterns of renal polyamine-oxidizing enzymes polyamine oxidase (PAO) and diamine oxidase (DAO) in male and female ICR mice were demonstrated. The effects of testosterone (10 micrograms/100g body weight) on renal PAO and DAO activities were also studied. The differences between sexes in both PAO and DAO activities were most clearly expressed in the immature kidney. At the age of 20 days PAO and DAO activities were 1.52 fold (p < 0.01) and 1.75 (p < 0.02) respectively higher i… Show more

“…The diamine oxidase (DAO) activity was detected by using the methods as described by Jotova et al (1999), Su et al (2005), Xing et al (2007) and Yang et al (2011). The reaction solutions (3.0 ml) contained 2.5 mL 0.1 M sodium phosphate buffer (pH 6.5), 0.1 mL crude enzyme extracts, 0.1 mL peroxidase (250 U mL − 1 ) and 0.2 mL 4-aminoantipyrine / N, N-dimethylaniline.…”

“…One of these two scenarios must clearly occur, because similar high levels of Spd and Spm were found in male and female kidneys. The presence of N 1 -SSAT and PAO in male and female kidneys (14) indicates that catabolism of Spd and Spm takes place and leads to Put production. However, it seems that the interconversion pathway is not sufficient to compensate for the lack of ODC activity and to supply enough Put to the female mouse kidney.…”

“…Enzymes involved in polyamine metabolism, such as ODC, SAMDC, and PAO, are known to be especially abundant in the male mouse kidney (7,12,14,22). ODC mRNA, protein, and enzyme activity have been clearly localized by various techniques in the renal cortex, particularly in the proximal convoluted tubule (PCT) of the male mouse (7,18,21,31).…”

“…Whilst body weight was lower in female double transgenic mice compared to SOD1 G93A mice, in males both SOD1 G93A and double transgenic animals showed a similar body mass ( Fig 4 ). This differential effect in females may reflect an endocrine effect, as testosterone is known to stimulate DAO activity in mouse kidney [ 28 ].…”
