Devising tissue ingrowth metrics: a contribution to the computational characterization of engineered soft tissue healing

Abstract: The paradigm shift brought about by the expansion of tissue engineering and regenerative medicine away from the use of biomaterials, currently questions the value of histopathologic methods in the evaluation of biological changes. To date, the available tools of evaluation are not fully consistent and satisfactory for these advanced therapies. We have developed a new, simple and inexpensive quantitative digital approach that provides key metrics for structural and compositional characterization of the regenera… Show more

“…[183] Picrosirius Red consists of elongated dye molecules that readily react with amino acid rich collagen molecules. [184] Thus, the dye enhances the natural birefringence of collagen by aligning itself in parallel with each collagen molecule. [185] Differences in the birefringence of con stituent molecules can be used to identify collagen in a noncol lagenous environment and to differentiate individual collagen types, albeit to a certain degree.…”

“…Picrosirius Red staining, for example, of collagenous tissues has been used in conjunction with polarized light microscope to detect fiber quantity and hue . Picrosirius Red consists of elongated dye molecules that readily react with amino acid‐rich collagen molecules . Thus, the dye enhances the natural birefringence of collagen by aligning itself in parallel with each collagen molecule .…”

“…Anisotropy in the red spectrum suggested the predominance of type I collagen in the green spectrum -type III collagen. 33,34 Histological slides of wound tissues were evaluated by the semi-quantitative analysis system described earlier. In each section, signs of inflammation (exudation, presence of bacterial colonies, inflammatory infiltration, microcirculation disorders) and regeneration (angiogenesis, fibroblast proliferation, volume, and maturity of granulation tissue) were assessed on a 5-point scale (0 -no, 4 -maximum intensity).…”
