Diagnosis of Nasopharyngeal Carcinoma by DNA Amplification of Tissue Obtained by Fine-Needle Aspiration

Abstract: In patients with suspected nasopharyngeal carcinoma, fine-needle aspiration can provide tissue for diagnosis by DNA amplification of EBV genomes. The presence of EBV in metastases from an occult primary tumor is predictive of the development of overt nasopharyngeal carcinoma.

“…DNA was extracted from the tumour tissues and blood leucocytes by methods previously described (Maniatis et al, 1989). EBV detection and typing by PCR For the detection and typing of EBV DNA in the tumour tissues, three previously described polymerase chain reaction (PCR) protocols were used with some modifications (Sample et al, 1990;Feinmesser et al, 1992;Lin et al, 1993). DNA from cell line B958, EBV-transformed human lymphocytes (American TIype Culture Collection), was used as positive control and double-distilled water as negative control.…”

“…The fusion of both imaging modalities in an integrated PET/CT machine should give better results. 14,[23][24][25] Furthermore, useful information can be obtained during a single seating and also time saving for the patient's convenience.…”

“…A fused integrated morphological and functional imaging modality of positron emission tomography / computed tomography (PET/CT) is another possible useful new tool to be utilized in the assessment of NPC patients like in other oncological patients. 14 This study was conducted in view to evaluate the role of fused PET/CT in the management of NPC patients.…”

“…RT-PCR products were electrophoretically separated. Where indicated, gels were blotted to Hybond-N ϩ membranes (Amersham Pharmacia Biotech) and hybridized with the following ␥-32 Pend-labeled reporter probes (23,24): ZNF EB and ZNF EB sv (ZNF EB 1858, 5Ј-CCATTGTGGAGAAGACAGTC-3Ј); EBNA1 (109194, 5Ј-TGAAT ACCACCAAGAAGGTG-3Ј); and ␤-glucuronidase (GUS 1405, 5Ј-CTACTACTTGAAGATGGTGATCG-3Ј). Photography of gels/blots was performed using a Chemi Imager (Alpha Innotech, San Leandro, CA).…”
