Dickkopf-1 regulates gastrulation movements by coordinated modulation of Wnt/βcatenin and Wnt/PCP activities, through interaction with the Dally-like homolog Knypek

Abstract: Dickkopf-1 (Dkk1) is a secreted protein that negatively modulates the Wnt/␤catenin pathway. Lack of Dkk1 function affects head formation in frog and mice, supporting the idea that Dkk1 acts as a "head inducer" during gastrulation. We show here that lack of Dkk1 function accelerates internalization and rostral progression of the mesendoderm and that gain of function slows down both internalization and convergence extension, indicating a novel role for Dkk1 in modulating these movements. The motility phenotype f… Show more

“…Heterooligomerization of Fz-LRP5/6 is sufficient for activating Wnt/ -catenin signaling as demonstrated elegantly by studies involving chimeric Fz-LRP5/6 and Fz-Dkk proteins [210][211][212]. Evidence suggests that LRP6, in addition to participating positively in Wnt/ -catenin signaling, may also be engaged in an inhibitory role incatenin independent Wnt signaling [213,214].…”

“…65,66 DKK1 and LRP6 are regulators of canonical Wnt signaling, but they may also induce noncanonical Wnt signaling pathways, 67,68 both being implicated in the pathology of PAH. 10,13,15,69,70 To date, we do not understand why carriers of mutations in BMPR2 and other genes (e.g., CAV1) develop PAH; however, a common underlying feature is decreased BMPR2 signaling activity.…”

“…55 Inhibition of canonical Wnt signaling might result in activation of non-canonical JNK signaling (see below for details), as shown in several studies. 56,57 NON-CANONICAL WNT PATHWAY Non-canonical Wnt signaling activities include Wnt/JNK/PCP pathway and Wnt/Ca 2+ pathway. Both of them are b-catenin-independent pathways and require heterotrimeric G proteins for Fz function.…”
