Differences in the intrinsic chondrogenic potential of equine umbilical cord matrix and cord blood mesenchymal stromal/stem cells for cartilage regeneration

Abstract: Umbilical cord blood mesenchymal stromal/stem cells (UCB-MSCs) and umbilical cord matrix MSCs (UCM-MSCs) have chondrogenic potential and are alternative sources to standard surgically derived bone marrow or adipose tissue collection for cartilage engineering. However, the majority of comparative studies explore neonatal MSCs potential only on ISCT benchmark assays accounting for some bias in the reproducibility between in vitro and in clinical studies. Therefore, we characterized equine UCB-MSCs and UCM-MSCs a… Show more

“…Briefly, cells displayed a conventional phenotype for equine MSCs (i.e. CD44+; CD29+; CD90+; MHC1+; MHC2-; CD45-; CD34-), differentiated in vitro into adipogenic and osteogenic cell lineages and expressed chondrogenic markers upon differentiation [30]. MSCs were frozen in 10% (v/v) DMSO and stored in liquid nitrogen.…”

“…While comparing the therapeutic potentials of other adult tissue- and UCB-derived MSCs, a similar pattern in the extent and level of chondrogenic differentiation capacity was demonstrated[ 58 , 59 ]. Moreover, similar increases in proteoglycans were detected by safranin O staining[ 14 ].…”

“…Therefore, Promerim ® may be useful during the two phases of the ACI procedure to increase the proliferation/amplification of cells and foster their synthesis during the differentiation/redifferentiation step, while inhibiting their senescence (Promerim ® 30). Furthermore, collagen hydrolysates may be of interest to enhance MSC chondrogenesis, all the more so if these hydrolysates are as active as some of the members of the TGF-β family [ 92 , 93 , 94 , 95 , 96 , 97 ].…”
