“…Most previous large‐animal models of transplantation have entailed the use of radiation to deplete endogenous spermatogenesis, 43‐45 as considerable injury to the haematopoietic system is provoked by the toxicity of alkylating agents; however, alkylsulfonate busulfan is favourable to the establishment of testicular recipient models for cellular transplantation in mice 46 and monkeys 47 . We therefore utilized the approach reported by Hermann et al to eliminate endogenous spermatogenesis while simultaneously counteracting the damage to the hematopoietic system.…”

“…Consequently, busulfan treatment is the most common method used to prepare recipients of spermatogonial stem cells transplantation [27,28] and to study spermatogonial stem cells kinetics and fertility recovery [29,30,31]. Busulfan can eliminate almost all endogenous germ cells in the recipient, creating an empty space in the spermatogonial stem cells niche; therefore, this drug has been used successfully to prepare recipients in mice [32,33], rats [34,35], pigs [36,37], dogs [38], hamsters [39], monkeys [40], and roosters [41].…”

“…Since PGCs are precursors of sperm, researchers suggested that roosters could be used as recipient for exogenous transfer of genetically-modified PGCs which may improve the germline transmission rate (Trefil et al, 2017 ). Chicken embryos and adult roosters were chemically or physically sterilized to create a surrogate for external PGC donors (Trefil et al, 2006 ; Nakamura et al, 2008 , 2010 ; Ghadimi et al, 2017 ). Nakamura and colleagues partially sterilized chicken embryos by injecting Busulfan into the yolk of fertile eggs before incubation; this led to a significant reduction of endogenous PGCs.…”
