Differential Adeno-Associated Virus Serotype-Specific Interaction Patterns with Synthetic Heparins and Other Glycans

Abstract: All currently identified primary receptors of adeno-associated virus (AAV) are glycans. Depending on the AAV serotype, these carbohydrates range from heparan sulfate proteoglycans (HSPG), through glycans with terminal ␣2-3 or ␣2-6 sialic acids, to terminal galactose moieties. Receptor identification has largely relied on binding to natural compounds, defined glycan-presenting cell lines, or enzyme-mediated glycan modifications. Here, we describe a comparative binding analysis of highly purified, fluorescent-dy… Show more

“…Glycans 141/143 and 316/318 differ in that the terminal SIA in one of the branches in glycan 316/318 is linked by ␣2,6 rather than ␣2,3 to galactose. Binding of AAV5 to these sulfonated and branched glycans was also recently reported elsewhere (28).…”

“…For AAV2 production, HEK 293 cells seeded at 30% confluence were transfected 24 h later with AAV plasmids and pHelper as described previously (18). AAV2 was purified from benzonase-treated, cleared freeze-thaw supernatants by one-step AVB Sepharose affinity chromatography and quantified by qPCR as described previously (19) with primers for AAV2 rep (Table 1). AAV2 infectious titers were determined by endpoint dilutions on Ad2-infected HeLa cells (20).…”

“…To date, 13 distinct human or nonhuman primate AAV serotypes have been isolated. [22][23][24] The primary receptors for 8 of these serotypes have been identified so far. Whereas the prototype AAV2, as well as AAV3 and AAV13, recognizes heparan sulfate proteoglycans, AAV1, AAV4, AAV5, and AAV6 bind Nlinked or O-linked sialic acids and AAV9 cell surface terminal galactose residues.…”
