Differential effects of estradiol on carotid artery inflammation when administered early versus late after surgical menopause

Abstract: Objective To determine the effects of estrogen therapy (ET) on carotid artery inflammation when initiated early and late relative to surgical menopause. Methods and Results Female cynomolgus macaques consuming atherogenic diets were ovariectomized and randomized to control or oral estradiol (E2, human equivalent dose of 1 mg/d micronized E2) initiated at 1 month (Early menopause, n=24) or 54 months, (Late menopause, n=40) post-ovariectomy. Treatment period was 8 months. Carotid artery expression of markers o… Show more

“…For cortisol measurements, a morning blood samples were taken within 5 min of capture and sedation with ketamine hydrochloride (10 mg/kg) and used with commercially available coated tube RIA kit from Diasource (Louvain-la-Neuve, Belgium). Total plasma cholesterol (TPC) and HDL were measured as previously described ( Sophonsritsuk et al, 2013 ). In brief, TPC and HDL were determined in the Wake Forest Primate Center Clinical Chemistry Laboratory using ACE cholesterol and ACE HDL-C reagents and ACE ALERA autoanalyzer from Alfa Wasserman Diagnostic Technologies (West Caldwell, NJ).…”

“…In the present study, we chose to address some of these issues in climacteric women. Menopause is associated with chronic inflammation and has major implications for CAD development [24, 25] and associated lipid disorders [26]. HPV incidence is also higher in this group [27] and might act synergistically with some of these factors to aggravate its cardiovascular risk.…”

“…30 Experimental studies in animals have also provided support for this association. 31,32 Although, the mechanism by which aging and longer periods of estrogen deprivation affect HRT responses is unclear, there are suggestions that estrogen has a moreproinflammatory effect with aging. 33 It is difficult to interpret the current study findings in context of the timing hypothesis, because a surrogate marker of atherosclerosis was used, and event rates were compared using a cross-sectional approach, but the findings suggest that, if HRT is initiated early and continued for longer than 15 years, an association with low CAC will remain.…”
