Differential Expression of over 60 Chromosomal Genes in Escherichia coli by Constitutive Expression of MarA

Abstract: In Escherichia coli, the MarA protein controls expression of multiple chromosomal genes affecting resistance to antibiotics and other environmental hazards. For a more-complete characterization of the mar regulon, duplicate macroarrays containing 4,290 open reading frames of the E. coli genome were hybridized to radiolabeled cDNA populations derived from mar-deleted and mar-expressing E. coli. Strains constitutively expressing MarA showed altered expression of more than 60 chromosomal genes: 76% showed increas… Show more

“…The fact that the induced RNA concentration reaches a steady plateau in mutant cells indicates that the AcrAB-TolC multidrug efflux system contributes to the observed pulsating dynamics of the inducible tet promoter in wild-type cells (30). It is also conceivable that the observed pulsating behavior in wild-type cells may be caused by periodic RNA degradation.…”

“…MarA, SoxS and Rob each regulate the expression of multiple genes (called the MarA, SoxS and Rob regulons) in E. coli (Barbosa & Levy, 2000;Bennik et al, 2000;Pomposiello et al, 2001), S. typhimurium and, by inference, other members of the Enterobacteriaceae. This regulation is achieved by the binding of the transcription factor to a degenerate sequence, called the 'marbox' or 'soxbox' (Martin & Rosner, 2003), which is located within the promoter of the regulated gene.…”

“…Sirvan a título orientativo las discrepancias observadas por 2 grupos de investigación en la aplicación de chips de E. coli del mismo origen comercial: al estudiar la expresión de genes regulados por MarA (relacionado con la resistencia múltiple a los antimicrobianos), un grupo observó la modulación de 88 genes 23 y el otro de sólo 62 24 , y sólo 21 de estos genes coincidían en ambos estudios.…”
