Differential oxidative damage and expression of stress defence regulons in culturable and non‐culturable Escherichia coli cells

Abstract: Potentially pathogenic bacteria, such as Escherichia coli and Vibrio cholerae, become non-culturable during stasis. The analysis of such cells has been hampered by difficulties in studying bacterial population heterogeneity. Using in situ detection of protein oxidation in single E. coli cells, and using a density-gradient centrifugation technique to separate culturable and non-culturable cells, we show that the proteins in non-culturable cells show increased and irreversible oxidative damage, which affects var… Show more

“…Oxidatively damaged proteins increase during aerobic carbon starvation (Nystrom, 2004;Dukan and Nystrom, 1999), and cells which fail to control the burden of oxidized proteins lose viability (Desnues et al, 2003). We thus speculate that the aerobically-driven increase in oxidized proteins may be mechanistically important for induction of the adaptive response in Lon deficient cells.…”

“…In addition, the data imply that cytokinesis during binary fission is also inherently asymmetrical. Indeed, previous to these studies it has been shown that cells of an E. coli population consist of two discrete populations with respect to damage: a damage-enriched and a damage-free population [33]. The damage-free cells remain reproductively competent in stationary phase, whereas the damage-enriched cells eventually become genetically dead (non-culturable).…”

“…To deal with oxidative stress, bacteria are known to evolve protective responses including the peroxidase-, catalase-, and superoxide dismutase (SOD)-mediated detoxification of ROS intermediates and repair of oxidative damage to bacterial DNA (Rosner and Storz, 1997;Desnues et al, 2003;Noor et al, 2009aNoor et al, , 2009bMurata et al, 2012;Munna et al, 2013;Nagamitsu et al, 2013). In addition, Escherichia coli produces cytoplasmic Mn-SOD (sodA) and Fe-SOD (sodB), which protect DNA and proteins from oxidation, while periplasmic Cu/Zn-SOD (sodC) protects the periplasmic and membrane constituents from exogenous O 2 - (Cabiscol et al, 2000;Moat et al, 2002;Rensing and Grass, 2003;Wang et al, 2007).…”
