Differential response of MCF7, MDA-MB-231, and MCF 10A cells to hyperthermia, silver nanoparticles and silver nanoparticle-induced photothermal therapy

Abstract: Ag NP are effective photothermal agents. A secondary benefit is the differential response of breast cancer cells to Ag NP-induced hyperthermia, due to increased intracellular silver content, compared to non-tumorigenic breast epithelial cells.

“…5, 25, 50, and 75 nm in diameter spherical AgNPs stabilized with polyvinylpyrrolidone (PVP; 89.3, 85, 76.5, 74% by mass, respectively), PVP and silica shelled triangular silver nanoplates, and PVP‐stabilized 15 nm gold nanoparticles (AuNPs; 91.6% PVP by mass) were purchased as dried nanopowders from nanoComposix, Inc. Free (ionic) silver represented less than 1% of the total nanoparticle silver content according the manufacturer's specifications. Chitosan‐coated AgNPs were synthesized according to previously published methods . Nanoparticles were dispersed at a concentration of 20 mg/mL (total nanoparticle weight) in phosphate buffered saline (PBS) (Invitrogen) by bath sonication, and then were diluted in cell culture medium to the final concentration listed in the figures prior to addition to the wells containing cells.…”

“…Silver nanoparticles are inherently highly effective against microorganisms by inhibiting bacterial growth on medical devices and burns, especially dependent upon their surface area [119][120][121]. They can be made more anisotropic to facilitate the shift of their plasmon resonance, and hence optical absorption, into the NIR, so that they can be used as photothermal agents [122]. Many NP designed to couple photothermal heating with delivery utilise classic antibiotics.…”

“…Despite the advantages of the use of AuNPs, some studies report the use of AgNPs in photothermal therapy against different types of cancer, including ovarian cancer [64]. AgNPs have photothermal and cytotoxic properties, which can be used to generate a synergistic effect [61,65,66], which leads to the death of cancer cells. The type of nanostructures that have frequently been used in the field of biomedicine includes nanospheres, nanowires, nanorods, nanoplates and nanocubes [2,22,25,50,56,74].…”
