Differentially expressed galactinol synthase(s) in chickpea are implicated in seed vigor and longevity by limiting the age induced ROS accumulation

Abstract: Galactinol synthase (GolS) catalyzes the first and rate limiting step of Raffinose Family Oligosaccharide (RFO) biosynthetic pathway, which is a highly specialized metabolic event in plants. Increased accumulation of galactinol and RFOs in seeds have been reported in few plant species, however their precise role in seed vigor and longevity remain elusive. In present study, we have shown that galactinol synthase activity as well as galactinol and raffinose content progressively increase as seed development proc… Show more

“…3B ). RFOs accumulate during seeds development is thought to be important for desiccation tolerance during seed maturation and longevity in dehydrated state 18 , 39 . Seeds of the AtRS4 and AtRS5 double mutant showed a total loss of RFOs and five days delayed germination phenotype in darkness, suggesting that RFOs also act as a galactose store in seeds and are necessary for rapid germination in the dark 40 .…”

“…There are some publications suggesting that galactinol positively regulates seed vigor in A. thaliana and some other plant species (Salvi et al, 2016;de Souza Vidigal et al, 2016). Galactinol was considered as a marker for seed storability in A. thaliana, cabbage, and tomato (de Souza Vidigal et al, 2016), and overexpression of the chickpea GALACTINOL SYNTHASE (CaGOLS) gene in A. thaliana increased the galactinol and raffinose content and enhanced seed vigor (Salvi et al, 2016). Since both galactinol and raffinose were increased by overexpression of the CaGOLS gene, it is not clear whether galactinol, raffinose, or both directly enhanced A. thaliana seed vigor.…”

“…In addition, galactinol and raffinose are substrates for synthesis of HDP RFOs, such as stachyose or verbascose. The change of stachyose or verbascose and their effects on A. thaliana seed vigor was not investigated in CaGOLS expressing A. thaliana (Salvi et al, 2016). This information is important because our evidence from overexpressing one or more genes whose products are involved in RFO biosynthesis suggest that the total amount of RFOs and the ratio of RFOs/ sucrose, but not galactinol, are responsible for A. thaliana seed vigor (Table 1 and Figure 6B).…”

“…Likewise, the abundance of HSP17.6I was positively correlated with seed performance in this species [74] and changes in enzyme activities in four Brassica species were proposed as indicators of seed deterioration [75]. Similarly, several studies have proposed addressed the use of novel seed quality markers, such as the production of methanol, ethanol or galactinol in stored seeds [11,12,[76][77][78]. However, the development of novel, equally reliable methods is always desirable.…”
